目的:观察刺络泻血疗法对高脂血症模型大鼠载脂蛋白CⅢ(ApoCⅢ)及脂蛋白酯酶(LPL)活性的影响,并探讨其作用机制。方法:将34只清洁级雄性SD大鼠分为空白组、模型对照组、立普妥组及刺络泻血组,每组8只,以高脂饲料喂养造模,造模持续28 d。第4周造模成功后,各造模组仍给予高脂饲料喂养,空白组不进行任何干预;模型组仅抓取按压处理,2次/周;立普妥组按照1.8 mg/(kg·d)的剂量给大鼠灌胃阿托伐他汀钙,2次/周;刺络泻血组选取大鼠委中穴及足三里穴穴位附近的血络,出血量为0.3~0.5 mL,至第8周。第9周腹主动脉取血,采用酶联免疫吸附法测定ApoCⅢ,采用光谱检测法测定LPL、TC、TG、HDL-C、LDL-C的含量。结果:实验结束后,与正常组比较,模型组ApoCⅢ、TC、LDL-C含量均显著增高,差异有统计学意义(P<0.05),LPL含量无明显变化(P>0.05);与模型组比较,刺络泻血组与立普妥组ApoCⅢ、TC、LDL-C含量显著降低,差异均有统计学意义(P<0.05),LPL含量无明显变化(P>0.05)。结论:刺络泻血疗法可以有效抑制ApoCⅢ活性,降低高脂血症大鼠TC、LDL-C水平,还不能认为对LPL有干预作用。 |

To observe the effects of pricking blood therapy on ApoCⅢ and LPL in hyperlipidemia rats and explore its mechanism. Methods:A total of 34 male SD rats were randomly divided into 4 groups:normal group, model group, lipitor group and blood pricking therapy group, with 8 in each group. Hyperlipidemia model was induced by high fat diet for 28 days. After successful modeling of 4 weeks, each group was still given high fat diet except control group. Rats in normal group didn't accept any interventions, while the model group was treated with grabbing and pressing, twice per week. The lipitor group was given atorvastatin calciumby intragastric administration with 1.8 mg/(kg·d) each time, twice per week. In the blood pricking therapy group, the superficial vein of Weizhong and Zusanli were selected for 0.3 to 0.5 mL bleeding until the eighth week. Blood was collected from the abdominal aorta on the 9th week, and ApoCⅢ was detected by enzyme-linked immunosorbent assay (ELISA). The levels of LPL, TC, TG, HDL-C and LDL-C were measured by spectrophotometry. Results:Comparing with the control group, the level of TC, LDL-C and ApoCⅢ of the model group were significantly increased, and the difference was statistically significant (P<0.05). No significant changes were seen in LPL (P>0.05). Compared with the model group, the level of TC, LDL-C and ApoC3 of the blood pricking therapy group and lipitor group decreased significantly, and the differences were statistically significant (P<0.05). No significant changes were seen in LPL (P>0.05). Conclusion:Blood-letting therapy can effectively inhibit the activity of ApoC III and reduce the levels of TC and LDL-C in hyperlipidemia rats, but it cannot be considered as an intervention to LPL. |
