      目的:对五倍子鞣质的提取工艺进行优化,为复方止血消炎软膏的制备工艺提供一定参考依据。方法:建立没食子酸的高效液相色谱分析方法,其色谱条件为:流动相:甲醇-乙腈-0.2%磷酸(4:11:85);柱温:30 ℃;检测波长:273 nm;流速:1 mL/min,进样量:10 μL。然后,以没食子酸提取率为指标,在单因素考察的基础之上,固定提取次数,以水的用量,提取时间,提取温度为因素,选用L9(34)正交表进行正交试验优化总鞣质的提取工艺。结果:建立的高效液相色谱分析方法的精密度、重复性、稳定性均符合药典要求,没食子酸在2.34~300 μg范围内进样量与色谱峰面积呈现良好的线性关系,其回归方程为:Y=29.955X+9.635 2,r=1,其平均加样回收率为101.88%。正交试验优选的鞣质最佳提取工艺为A3B1C3,即在80 ℃水浴中,用12倍量的水作为溶剂,提取2次,1.5 h/次。结论:本文所建立的高效液相色谱法色谱分析方法准确,稳定,可用于没食子酸的含量测定;正交试验优选的五倍子提取工艺稳定可靠,适用于总鞣质的提取。
English Summary:
      To optimize the extraction process of Galla Chinensis tannin,and to provide a reference for the preparation of compound Zhixue Xiaoyan ointment.Methods:HPLC method for the analysis of gallic acid was established.The chromatographic conditions were as follows:mobile phase:methanol:acetonitrile:0.2% phosphate 4:11:85; column temperature:30 ℃; detection wavelength:273 nm; flow rate:1 mL/min,injection amount:10 μL.Then,the extraction rate of gallic acid was taken as the index,the extraction times were fixed on the basis of single factor investigation,and the orthogonal experiment was carried out to optimize the extraction process of total tannin by orthogonal test with water dosage,extraction time and extraction temperature as factors.The L9(34)orthogonal table was used to carry out the orthogonal test to optimize the extraction process of total tannin.Results:The precision,repeatability and stability of the HPLC method were all in line with the requirements of pharmacopoeia.The linear relationship between the concentration of gallic acid in the range of 2.34~300 μg and the peak area was good.The regression equation was Y=29.955X+9.6352,the correlation coefficient r=1,and the average recovery rate was 101.88%.The optimum extraction process of tannin by orthogonal test was A3B1C3,that is,in 80 ℃ water bath,12 times of water was used as solvent to extract tannin twice for 1.5 hours each time.Conclusion:The HPLC method established in this paper is accurate and stable,and can be used for the determination of gallic acid,and the optimum extraction process of Galla Chinensis by orthogonal test is stable and reliable,which is suitable for the extraction of total tannins.
