      目的:评价确证天麻硫熏标志物(p-hydroxybenzyl hydrogen sulfite)和硫熏天麻水提物的细胞毒性。方法:采用CCK-8试验方法,考察天麻硫熏标志物和水提物不同剂量下人肾皮质近曲小管上皮细胞、人正常肝细胞和大鼠肾上腺嗜铬细胞瘤细胞的生长情况,计算相应的细胞活力,并根据相对增殖率进行细胞毒性评级。结果:硫熏标志物p-hydroxybenzyl hydrogen sulfite对于人体2种细胞的生长均表现出良好的促进作用,且参照《美国药典》的毒性分级法,得出该标志物对大鼠肾上腺嗜铬细胞瘤细胞的安全剂量范围为0~1 000 μmol/L。另外发现,硫熏天麻水提物对人体正常细胞的细胞活力影响不大,与硫熏前的差异无统计学意义(P>0.05),并得出该标志物对大鼠肾上腺嗜铬细胞瘤细胞的安全剂量范围为0~2.5 μg/mL。结论:在正常的浓度范围内,硫熏标志物p-hydroxybenzyl hydrogen sulfite和硫熏天麻提取物均无细胞毒性,对细胞活力的影响不大。
English Summary:
      To evaluate the cytotoxicity of the water extract and p-hydroxybenzyl hydrogen sulfite in sulfur-fumigated Gastrodia Rhizoma.Methods:The CCK-8 test method was used to investigate the growth of human renal cortical proximal tubular epithelial cells,human normal hepatocytes and rat adrenal pheochromocytoma cells at different doses of sulfur-fumigated markers and water extracts for Gastrodia Rhizoma.And the vitality and cytotoxicity ratings based on relative proliferation rates was also evaluated.Results:The p-hydroxybenzyl hydrogen sulfite had a good promoting effect on the growth of both human cells,and the toxicity of the United States Pharmacopoeia was used to obtain the marker for rat adrenal pheochromocytoma cells.The safe dose range was from 0 to 1000 μmol/L.In addition,it was found that the water extract of sulfur-fumigated Gastrodia Rhizoma had little effect on the cell viability of normal human cells,and there was no significant difference(P>0.05)before and after of sulfur-fumigating Gastrodia Rhizoma,and the safe dose range of the marker on rat adrenal pheochromocytoma cells was 0~2.5 μg/mL.Conclusion:In the normal concentration range,the p-hydroxybenzyl hydrogen sulfite and water extract of sulfur-fumigated Gastrodia Rhizoma were not cytotoxic,and had little effect on cell viability.
