      目的:观察六神丸对放射性肺炎大鼠的治疗作用,对其血清中IL-1β、IL-6、TNF-α和TGF-β1进行检测,探究六神丸对放射性肺炎的治疗作用及可能的分子机制。方法:将40只雄性SD大鼠随机分为正常组、模型组、激素组、六神丸组,采用直线加速器对除正常组外的3组大鼠进行全胸单次照射,剂量为16 Gy。照射24 h后各组进行给药。照射后第4周,处死动物。取全肺称重计算肺系数;进行HE染色。采用Luminex法检测大鼠血清中细胞因子的表达。结果:与正常组比较,照射后大鼠肺系数显著升高(P<0.05),使用激素或六神丸干预后,大鼠肺系数较模型组低,差异有统计学意义(P<0.05,P<0.01)。HE结果显示激素组、六神组大鼠肺组织出现充血、水肿、渗出等炎性病理学改变均较模型组有改善,炎细胞浸润也不同程度减轻。六神丸可显著下调大鼠血清中IL-1β、TNF-α炎性反应因子的表达(P<0.05,P<0.01)。结论:放射性肺炎大鼠血清中炎性因子明显增加的增加,六神丸可有效减轻放射性肺炎大鼠局部肺损伤的程度,其机制可能与下调血清中TNF-α及IL-1β相关。
English Summary:
      To observe the effects of Liushen Pills on IL-1β, IL-6, TNF-α and TGF-β1 in the serum of rats with radiation pneumonitis(RP), and explore the therapeutic effect of Liushen Pills(LS)on RP and the possible molecular mechanism.Methods:A total of 40 male SD rats were divided into a normal control(NC)group, a model group, a prednisone acetate(PA)group, a LS group, randomly.Linear accelerator irradiated in the 3 groups except the NC group for single chest with the dose of 16Gy in rats.The rats in each group were given the drug by perfusion 24 hours after irradiation.After 4 weeks, the rats were sacrificed.The lung coefficient was calculated by weighing the lungs.The pathological changes in the lungs were observed by hematoxylin-eosin staining.The expressions of cytokines in serum of rats were detected by Luminex.Results:Compared with the NC group rats, the lung coefficient of model rats were significantly increased(P<0.05), while the lung coefficient of PA-treated and LS-treated rats were markedly decreased compared with the model rats, and the difference was statistically significant(P<0.05, P<0.01).The HE results showed that the PA-treated, LS-treated rats inflammatory lesions such as congestion, edema, and exudation were lighten than model rats in lung tissues.Liushen Pills could significantly decreased the expressions of IL-1β, TNF-α in the serum of rats(P<0.05, P<0.01).Conclusion:The proinflammatory cytokines in the serum of rats with RP were increased.Liushen Pills can effectively reduce lung injury in rats with RP, which may be related to the down-regulation of TNF-α and IL-1β in serum.
