      目的:探讨骨伤三号方对骨质疏松压缩性骨折模型鼠的治疗效果,同时探讨其对OPG/RANKL/核因子-κB信号通路的影响。方法:将30只雌性Sprague Dawley(SD)鼠随机分为2组,空白组(10只)及造模组(20只),20只造模术首先接受双侧卵巢切除术,术后4周再采用剪断L1-5节段椎体前缘以模拟骨质疏松压缩性骨折模型,共有18只大鼠成模,将其随机分为模型组及干预组,每组9只。干预组大鼠找造模成功后接受骨伤三号方灌胃,1次/d。空白组及模型组接受等量生理盐水灌胃,1次/d。各组均干预4周。干预结束后比较各组大鼠生物力学变化,用HE染色法观察各组大鼠骨痂形态结构变化,酶联免疫吸附试验检测各组血清OPG、核因子-κB水平变化;Western Blotting检测骨折组织RANK水平变化。结果:与空白组比较,造模组大鼠弹性模量、最大载荷降低,差异有统计学意义(P<0.05);与模型组比较,干预组弹性模量、最大载荷显著升高,差异有统计学意义(P<0.05)。HE染色显示干预4周后空白组可见成熟骨小梁形成,骨质疏松性骨折组骨痂邻近原骨小梁细小松散,向骨性骨痂转化过程明显延迟;干预组形态介于正常骨折组和骨质疏松性骨折组之间。与空白组比较,模型组及干预组大鼠OPG明显下降,核因子-κB、RANK明显升高,经过中药干预后大鼠OPG水平上调,核因子-κB、RANK表达减少,差异有统计学意义(P<0.05)。结论:骨伤三号方可能通过OPG/RANKL/核因子-κB信号通路加速骨质疏松压缩性骨折的愈合。
English Summary:
      To explore efficacy of Gushang No.3 Formula in osteoporotic compression fracture model rats and its effects on the OPG/RANKL/NF-κB signaling pathway.Methods:Thirty female Sprague Dawley(SD)rats were randomly divided into 2 groups:blank group(n=10)and modeling group(n=20).Firstly,the 20 rats received bilateral oophorectomy.Four weeks after the operation,the anterior edge of L1-L5 segment was cut to simulate the model of osteoporotic compression fracture.A total of 18 rats were successfully modeled,and randomly divided into model group(n=9)and intervention group(n=9).After the successful establishment of the model,the rats in the intervention group were given Gushang No.3 Formula by intragastric administration once a day.The blank group and the model group were given the same amount of normal saline once a day.All the groups were intervened for 4 weeks.After the intervention,biomechanical changes of rats in each group were compared.Morphological and structural changes of callus in each group were observed by HE staining,concentrations of OPG and NF-κB in serum in each group were detected by ELISA,and Western Blotting was used to detect changes of RANK level in fracture tissue.Results:1)Compared with the blank group,the elastic modulus and maximum load in the modeling group were decreased(P<0.05),and the elastic modulus and maximum load in the intervention group were significantly higher than those in the model group(P<0.01).2)The HE staining showed that mature bone trabeculae could be seen in the blank group after 4 weeks of the intervention.The callus in the osteoporotic fracture group was fine and loose,and the transformation to the bone callus was significantly delayed.The morphology in the intervention group was between that in the normal fracture group and that in the osteoporotic fracture group.3)Compared with the blank group,the OPG in the model group and the intervention group was decreased significantly,and the NF-κB and RANK were increased significantly.After the intervention of Chinese materia medica,the level of OPG was up-regulated and the expressions of NF-κB and RANK were decreased.The difference was statistically significant(P<0.05).Conclusion:Gushang No.3 Formula may accelerate healing of osteoporotic compression fracture through the OPG/RANKL/NF-κB signal pathway.
