      目的:研究归芪活血胶囊对颈椎病模型大鼠软骨细胞白细胞介素-6(IL-6)、白细胞介素-8(IL-8)及MKK3/6-p38MAPK信号通路的影响。方法:选取60只SD级大鼠作为研究对象,将其以随机抽签法等分成假手术组、模型组、归芪活血胶囊组。其中模型组与归芪活血胶囊组参考颈椎间盘退变模型,假手术组仅切开皮肤,随后缝合。模型组不予以任何干预,归芪活血胶囊组则予以归芪活血胶囊灌胃治疗。干预4周后,通过甲苯胺蓝实施组织染色,以免疫组织化学法检测软骨细胞IL-6、IL-8含量,同时以Weatern Blot法检测MKK3/6以及p38MAPK表达水平。结果:假手术组、归芪活血胶囊组大鼠颈椎间盘软骨细胞IL-6水平、IL-8水平均显著低于模型组,差异均有统计学意义(P<0.05)。假手术组、归芪活血胶囊组大鼠颈椎间盘软骨细胞MKK3/6水平、p38MAPK水平相较于模型组明显更低(均P<0.05)。经Pearson相关性分析可得:颈椎病模型大鼠软骨组织IL-6、IL-8水平与MKK3/6、p38MAPK表达水平比较,差异均有统计学意义(均P<0.05)。结论:归芪活血胶囊的应用可显著降低颈椎病模型大鼠软骨细胞IL-6、IL-8含量,同时可下调软骨细胞MKK3/6以及p38MAPK蛋白表达水平,其主要机制可能是通过调节MKK3/6-p38MAPK信号通路,进一步下调IL-6、IL-8含量,从而达到治疗颈椎病的目的。
English Summary:
      To study the effects of Guiqi Huoxue Capsule on interleukin-6(IL-6),interleukin-8(IL-8)and mkk3/6-p38mapk signaling pathway in cervical spondylosis model rats.Methods:A total of 60 SD rats were selected as the research objects,and they were randomly divided into a sham operation group,a model group and a Guiqi huoxue Capsule group.The model group and the Guiqi Huoxue Capsule group referred to the cervical disc degeneration model,while the sham operation group only cut the skin and sutured it afterwards.The model group was not given any intervention,while the Guiqi huoxue Capsule group was given gavage to the treatment group.After 4 weeks of intervention,tissues were stained with toluidine blue,and IL-6 and IL-8 contents in chondrocytes were detected by immunohistochemistry,and mkk3/6 and p38MAPK expression levels were detected by Weatern Blot.Results:The IL-6 levels of cervical intervertebral disc chondrocytes in the sham operation group and the Guiqi Huoxue Capsule group were which were significantly lower than those in the model group,with significant differences(P<0.05).The levels of MKK3/6 and p38MAPK of chondrocytes in cervical intervertebral discs of rats in the sham operation group and Guiqi Huoxue Capsule group were significantly lower than those in the model group(both P<0.05).Pearson correlation analysis showed that the levels of IL-6 and IL-8 in cartilage tissue of cervical spondylosis model rats were significantly different from the expression levels of MKK3/6 and p38MAPK(all P<0.05).Conclusion:The application of Guiqi Huoxue Capsule can significantly reduce the contents of IL-6 and IL-8 in chondrocytes of cervical spondylosis model rats,and down-regulate the expression levels of MKK3/6 and p38MAPK protein in chondrocytes.The main mechanism may be to further down-regulate the contents of IL-6 and IL-8 by regulating the MKK3/6-p38mapk signaling pathway,so as to achieve the purpose of treating cervical spondylosis.
