      目的:观察松花粉总黄酮对急性化学性肝损伤模型小鼠的作用机制。方法:60只ICR小鼠随机分为空白组、模型组及实验组,每组20只。其中模型组及实验组制备小鼠急性肝损伤模型,模型制备成功后实验组每日灌胃松花粉总黄酮,空白组及模型组灌胃同等剂量的生理盐水,连续干预10 d。比较3组肝脏病理变化,外周血谷丙转氨酶(ALT)、谷草转氨酶(AST)的活性以及PI3K-AKT信号通路标志性蛋白水平变化。结果:1)空白组小鼠肝脏形态正常,模型组小鼠肝脏出现明显病理改变,实验组小鼠肝脏细胞空泡及炎性反应侵润现象较模型组减轻,肝脏细胞轻微坏死。2)与空白组比较,模型组外周血ALT、AST及Bax浓度均明显上调(P<0.05),Bcl-2表达下降(P<0.05);实验组上述指标明显改善。3)模型组肝脏组织的PI3K、p-AKT水平明显下降,干预后小鼠上述指标表达明显上调,与模型组比较差异有统计学意义(P<0.05)。结论:松花粉总黄酮对化学性肝损伤有保护效应,其作用机制可能与介导PI3K-AKT信号通路有关。
English Summary:
      To observe the mechanism of total flavonoids of pine pollen on acute chemical liver injury model mice. Methods:A total of 60 Institute of Cancer Research( ICR) mice were randomly divided into blank group (n=20), model group (n=20) and experimental group (n=20). The mouse model of acute liver injury was made in the model group and the experimental group. After the successful preparation of the model, the experimental group was intragastrically infused with total flavonoids of pine pollen every day, and the blank group and the model group were intragastrically infused with the same dose of normal saline for 10 days. The pathological changes of liver, the activities of glutamic pyruvic transaminase (ALT) and glutamic oxaloacetic transaminase (AST) in peripheral blood and the levels of marker proteins in PI3K-AKT signal pathway were compared among the three groups. Results:1) the liver morphology of mice in the blank group was normal, and there were obvious pathological changes in the model group. The vacuolation and inflammatory infiltration of liver cells in the experimental group were less than those in the model group, and the liver cells were slightly necrotic. 2) compared with the blank group, the concentrations of ALT, AST and Bax in the model group were significantly increased, and the expression of Bcl-2 was decreased in the model group, and the above-mentioned indexes were significantly improved in the experimental group. 3) the levels of PI3K and p-AKT in the liver tissue of the model group decreased significantly, and the expression of the above-mentioned indexes increased significantly after intervention in the experimental group, which was significantly different from that of the model group (P<0. 05). Conclusion:Total flavonoids of pine pollen have protective effect on chemical liver injury, and its mechanism may be related to the mediation of PI3K-AKT signal pathway.
