      目的:观察补肾助孕方对肾虚薄型大鼠子宫内膜容受性的影响,探讨中药的作用机制。方法:雌性SD大鼠30只,随机分为对照组10只,造模组20只,通过羟基脲灌胃建立肾虚合并着床障碍型子宫内膜大鼠模型。随后将20只造模组大鼠随机分为模型组和补肾助孕方组,每组10只。对照组、模型组均灌服蒸馏水10 mL/kg;补肾助孕方组灌服中药0.3 mg/kg,均1次/d,持续3个动情周期(15 d)。在第3个动情期后处死大鼠,酶联免疫吸附法检测动脉血中雌二醇、孕酮的水平。取子宫标本,光镜下观察子宫内膜形态,测量子宫内膜厚度、子宫间质厚度、子宫腺体数、子宫血管数。免疫组化法检测各组内膜组织中整合素β3和白血病抑制因子的表达。结果:与正常对照组比较,模型组大鼠动脉血雌二醇、孕酮的水平减少;子宫内膜腺体稀少,上皮变薄,间质致密,血管少;子宫内膜厚度、子宫间质厚度、子宫腺体数、子宫血管数均减少;整合素β3和白血病抑制因子的表达减少。补肾助孕方组较之模型组,有明显的改善。结论:补肾助孕方可改善肾虚合并着床障碍型状态大鼠子宫内膜的容受性,可能与提高血中激素水平,增加内膜中整合素aβ3和白血病抑制因子的表达有关。
English Summary:
      To observe the effects of Bushen Zhuyun Formula on the endometrial receptivity of rat model with kidney deficiency and thinner endometrial, for exploring the possible mechanism of the Chinese medicine. Methods:A total of 30 female SD rats were randomly divided into a control group (10 rats) and a model group (20 rats). Rats model with kidney deficiency and embryo implantation dysfunction were established by intragastric administration of hydroxyurea. Then 20 rats in the modeling group were randomized into a model group, Bushen Zhuyun group with 10 in each.The control group and the model group was given oral administration of distilled water 10 ml/kg; the Bushen Zhuyun Formula group was given Chinese medicine 0.3 mg/kg, respectively. All these rats were intervened once daily, and the intervention lasted for 3 consecutive estrous cycles (15 days). Rats were sacrificed after the third estrus period. Enzyme-linked immunosorbent assay was used to detect the levels of estradiol and progesterone in arterial blood. Take a specimen of the uterus, observe the morphology of the endometrium under a light microscope, measure the thickness of the endometrium, the thickness of the interstitium, the number of uterine glands, and the number of uterine blood vessels. Immunohistochemical method was used to detect the expression of integrin β3 and leukemia inhibitory factor in the inner membrane tissue of each group. Results:Compared with the normal control group, the arterial blood estradiol and progesterone levels of rats in the model group were reduced; the endometrial glands were sparse, the epithelium became thin, the interstitium was dense, and the blood vessels were less; the thickness of the endometrium, the thickness of the interstitium, the number of uterine glands and uterine blood vessels decreased; the expression of integrin β3 and leukemia inhibitory factor decreased. Compared with the model group, the Bushen Zhuyun Formula group has obvious improvement. Conclusion:Bushen Zhuyun Formula can improve the endometrial receptivity of rat model with kidney deficiency and embryo implantation dysfunction, and the mechanism is related to increasing hormone levels in the serum and ?increasing the expressions of integirn β3 and leukemia inhibitory factor (LIF) in the endometrium.
