      目的:挖掘谷晓红教授治疗小儿急性上呼吸道感染的组方用药规律,分析组方可能存在的药物靶标及相互作用,探索核心药物的作用机制。方法:收集谷晓红教授医案数据库中小儿急性上呼吸道感染的医案,采用中医传承辅助平台软件分析药物间的关联规则,挖掘核心药物及新方组合;运用SPSS软件对药物的性味归经进行系统聚类分析;运用TCMSP、String及KEGG数据库分析新方组合各药物靶标间相互作用关系及相关通路。结果:在筛选出的164首处方中,挖掘出2个核心药物组合和2首新方组合,总结出性味归经相近的6个高频药群,得到新方组合活性成分195个,相关药物靶标1 406个,急性上呼吸道感染靶标31 312个,药物-疾病交集靶标1 385个,其中核心靶标300个。这些靶标构成的网络中共存在相互作用4 430项、相关通路25条(P<0.001),其中与呼吸系统关系最密切的是细胞凋亡、T细胞受体信号通路、MAPK信号通路、Toll样受体信号通路,新方组合核心靶标52个。结论:谷晓红教授治疗小儿急性上呼吸道感染以辛凉透表、和胃化湿法为主,其机制可能是通过调控细胞凋亡,调控炎性反应及免疫应答等多靶标复杂通路的共同干预实现。
English Summary:
      To explore the medication rule of Professor GU Xiaohong's treatment of pediatric acute upper respiratory infection(AURI),to analyze the interactions of drug targets in the component,and to explore the core mechanism of drugs.Methods:Cases of AURI from Professor GU Xiaohong's medical record database were collected,and Traditional Chinese Medicine Inheritance Support System(TCMISS)was used to analyze the association rules between drugs,core drugs and new component were mined.SPSS software was used to carry out systematic clustering analysis on the nature and flavor and channel tropism of drugs; TCMSP,String and KEGG database was used to analyze the interaction between the drug targets of the new prescription combination and related pathways.Results:A total of 164 prescriptions were selected,2 core drug combinations and 2 new prescriptions were explored.6 high-frequency drug groups with similar nature and flavor and channel tropism were summarized,and 195 active ingredients of the new prescription combination were obtained,with 1 406 related drug targets,and 31 312 targets for acute upper respiratory infections.The matching targets of drug-disease was 1 385,of which 300 were core targets.There were 4 430 interactions and 25 related pathways(P<0.001)in the network composed of these targets.Among them,the most closely related to the respiratory system were apoptosis,T cell receptor signaling pathway,MAPK signaling pathway,and Toll-like receptor signaling pathway.Xinfang combination 52 core targets Conclusion:Professor GU Xiaohong generally used principle of resolving exterior with pungent and cool natured drugs,regulating stomach and dissipating dampness to treat AURI.The mechanism may be achieved by co-intervention of multiple target complex pathways,such as regulating apoptosis,inflammatory response and immune response.
