      目的:研究复方511联合电针对小鼠吗啡条件性位置偏爱(CPP)和行为敏化的影响。方法:实验1,30只雄性C57BL/6J小鼠用随机数字表法随机分为对照组、模型组、复方511低、中、高剂量组,每组6只,采用CPP实验观察低、中、高剂量复方511对小鼠吗啡CPP偏爱分数及总活动度和伴药箱穿梭次数的影响;实验2,30只雄性C57BL/6J小鼠用随机数字表法随机分为对照组、模型组、复方511组、电针组、针药组,每组6只,采用CPP实验观察复方511、电针、复方511联合电针对小鼠吗啡CPP偏爱分数及总活动度和伴药箱穿梭次数的影响。结果:1)实验1,与对照组比较,模型组小鼠的偏爱分数显著升高(P<0.000 1),而低、中、高剂量复方511均能降低小鼠偏爱分数,但仅中剂量的差异有统计学意义(P<0.05)。与对照组小鼠比较,吗啡给药后小鼠的总活动路程和伴药箱穿梭次数较训练前明显减少(P<0.01),而施用中剂量复方511能显著提高减少的总路程和伴药箱穿梭次数(P<0.05)。2)实验2,与对照组比较,模型组小鼠的偏爱分数显著增加(P<0.000 1),复方511、电针、针药结合均能有效降低偏爱分数(P<0.05,P<0.05,P<0.000 1),且针药结合的作用优于单独使用复方511和电针(P<0.05)。与对照组比较,训练后模型组小鼠的总活动路程和伴药箱穿梭次数较训练前明显减少(P<0.000 1),而复方511、电针、针药结合均能显著提高小鼠吗啡给药后的总活动度和伴药箱穿梭次数(P<0.05)。结论:复方511联合电针能有效改善小鼠吗啡诱导的CPP和行为敏化。
English Summary:
      To observe the effects of compound 511 combined with electro-acupuncture on morphine induced-conditioned place preference(CPP)and behavioral sensitization in mice.Methods:A total of 30 male C57BL/6J mice were randomly divided into control group,model group,compound 511 low,medium,and high dose groups by random number table method with 6 mice in each group.The CPP experiment was used to observe the effects of low,medium and high dose of compound 511 in mice on morphine CPP preference score,total movement distance and the number of shuttles in the drug-paired box; 30 male C57BL/6J mice were randomly divided into the control group,the model group,the compound 511 group,the electroacupuncture group,and compound 511 combined electro-acupuncture group by the random number table method with 6 mice in each group.The CPP experiment was used to observe the effects of compound 511,electroacupuncture,compound 511 combined with electroacupuncture in mice on morphine CPP preference score,total movement distance and the number of shuttles in the drug-paired box.Results:1)Compared with the control group,the preference score of mice in the model group increased significantly(P<0.000 1).Low,medium and high doses of compound 511 could reduce the preference score,but only the difference of middle dose was statistically significant(P<0.05).Compared with the control group,the total movement distance and the number of shuttles in the drug-paired box in the model group were significantly reduced than pre-training period(P<0.01).The middle-dose compound 511 could significantly increase the total movement distance and the number of shuttles in the drug-paired box(P<0.05).2)Compared with the control group,the preference score of mice in the model group increased significantly(P<0.000 1).Compound 511,electroacupuncture,and compound 511 combined with electroacupuncture could significantly reduce the preference score(P<0.05,P<0.05,P<0.000 1),and the combination of electroacupuncture and compound 511 was better than using compound 511 and electroacupuncture alone(P<0.05).Compared with the control group,the total movement distance and the number of shuttles in the drug-paired box of the model group after training were significantly reduced compared with pre-training(P<0.000 1).Compared with the model group,the mice in the compound 511 group,electroacupuncture group,and acupuncture group had significantly more total movements and number of shuttles in the drug-paired box(P<0.05).Conclusion:Compound 511 combined with electroacupuncture can effectively inhibit morphine-induced CPP and behavioral sensitization in mice.
