      目的:探讨生姜、干姜、炮姜姜辣素部位在脾胃虚寒模型大鼠体内组织分布与归经的关系。方法:将雄性SD大鼠随机分为正常对照、生姜姜辣素、干姜姜辣素、炮姜姜辣素组;正常对照组大鼠灌胃去离子水,其余各组大鼠灌胃4 ℃米醋1 mL/100 g,连续10 d制备大鼠脾胃虚寒模型。造模第5天开始给药,连续给药6 d。各给药组于末次给药后不同时间点腹主动脉取血,并立即处死采集心、肝、脾、肺、肾、胃、大肠、小肠等组织,用UPLC-MS检测姜辣素6-姜酚、8-姜酚、10-姜酚、姜酮4种成分在大鼠各组织中的含量。采用DAS3.0软件分析药动学参数,对4种姜辣素成分在各组织中的AUC(0-t)值按从小到大的顺序依次赋值1,2,3,4,5,6,7,8,检测不到者赋值为0,计算各成分在各组织中AUC(0-t)的赋值总和并按大小排序,赋值总和大者表明选择性高,以此认识姜辣素类成分在体内的组织分布与归经的相关性。结果:4种成分在组织中AUC(0-t)赋值总和的排序结果为,生姜姜辣素:胃>大肠>小肠=肺>肾>肝>心>脾;干姜姜辣素:胃>小肠=大肠=肝>肺>脾>肾>心;炮姜姜辣素:胃>小肠>大肠>肝>肺>脾>心>肾。结论:生姜姜辣素组织分布与生姜归肺、脾、胃经基本一致;干姜姜辣素组织分布与干姜归脾、胃、肾、心、肺的认识基本一致;炮姜姜辣素组织分布与炮姜归脾、胃、肾经的认识基本一致。
English Summary:
      To investigate the relationship between tissue distribution and meridian tropism of gingerols from fresh,dried and baked ginger in rats with spleen-stomach deficiency cold model rats.Methods:SD male rats were randomly divided into control group,fresh gingerols group,dried gingerols group,and baked gingerols group.Rats in control group were given deionized water,while rats in other groups were given rice vinegar 1 mL/100 g at 4 ℃ for 10 consecutive days to prepare spleen-stomach deficiency cold model.The drug was administered at the 5th day after modeling,and 6 days in a row.The abdominal aorta blood was collected at different time points after the last administration of gingerols in each group,and the heart,liver,spleen,lung,kidney,stomach,large intestine,small intestine and other tissues were executed immediately.The content of 6-gingerol,8-gingerol,10-gingerol and zingiberone in the rat tissues were detected by UPLC-MS.Pharmacokinetic parameters were analyzed by DAS3.0 software.The AUC(0-t)values of four gingerols components in different tissues were sorted.The AUC(0-t)values were assigned to 1,2,3,4,5,6,7,8 in order from small to large,and those undetected were assigned to 0.The total value of AUC(0-t)in each tissue was calculated and sorted according to size.The higher the total value of AUC(0-t)was,the higher the selectivity was,so as to understand the correlation between the tissue distribution of gingerols and meridian tropism in vivo.Results:The total AUC(0-t)values of the four components in tissues were as follows:fresh ginger:stomach> large intestine> small intestine=lung> kidney> liver> heart> spleen; dried ginger:stomach> small intestine=large intestine=liver> lung> spleen> heart; baked ginger:stomach> small intestine> large intestine> liver> lung> spleen> heart.Conclusion:The distribution of fresh gingerols in tissues is basically the same as the meridian tropism of fresh ginger:the lung,spleen and stomach; the distribution of dried gingerols in tissues is basically the same as the meridian tropism of dried ginger:the spleen,stomach,kidney,heart and lung; the distribution of baked gingerols in tissues is basically the same as the meridian tropism of baked ginger:the spleen,stomach and kidney.
