      目的:通过网络药理学方法探讨经方真武汤治疗糖尿病肾病(DKD)的核心作用靶标基因以及潜在通路机制,旨在指导临床用药同时为更深入的实验研究提供依据。方法:通过TCMSP数据库检索真武汤所含药物的有效成分、潜在作用靶点,在Genecards、OMIM、DisGeNET数据库筛选与糖尿病肾病有关的疾病靶点,借助Cytoscape3.7.2绘制并构建“药物-成分-靶点-疾病”可视化交互网络。利用STRING平台绘制关键靶点PPI网络,在David数据库里针对关键靶点蛋白开展GO基因功能、KEGG信号通路富集分析。结果:检索查筛出真武汤中有效活性成分59个、潜在靶点108个,其中涉及DKD的关键靶点47个,从蛋白互作网络中分析发现TNF、ATK1、JUN、RELA、IL6、MAPK8、CASP8、NOS2可能是真武汤治疗糖尿病肾病的核心靶点。GO富集分析得到180个条目,主要涉及细胞增殖、代谢、凋亡及脂肪细胞因子、葡萄糖稳态等。KEGG信号通路富集分析得到94条信号通路,主要涉及TNF信号通路、Toll样受体信号通路、HIF-1信号通路、FoxO信号通路等。结论:本研究初步验证真武汤治疗糖尿病肾病的作用机制涉及多成分、多靶点、多信号通路,给予真武汤的临床应用及更深层次对糖尿病肾病的研究以一定理论依据。
English Summary:
      Through network pharmacology methods, the core target genes and potential pathway mechanisms of Jingfang Zhenwu Decoction in the treatment of diabetic nephropathy (DKD) are explored, aiming to guide clinical medication and provide basis for more in-depth experimental research. Methods:The active ingredients and potential targets of the drugs contained in Zhenwu Decoction were searched through the TCMSP database, and disease targets related to diabetic nephropathy were screened in the Genecards, OMIM, and DisGeNET databases. With the help of Cytoscape 3.7.2, the visualized interactive network of "drug-component-target-" was drawn and constructed. The STRING platform was used to draw the key target of PPI network, and carry out the GO gene function and KEGG signal pathway enrichment analysis for the key target protein in the David database. Results:A total of 59 effective active components combined with 108 potential targets were searched from Zhenwu Decoction, of which 47 key targets involved in DKD. TNF, ATK1, JUN, RELA, IL6, MAPK8, CASP8 and NOS2 may be the core targets of Zhenwu Decoction in the treatment of diabetic kidney disease. GO enrichment analysis yielded 180 entries, mainly involving cell proliferation, metabolism, apoptosis and adipocytokines, and glucose homeostasis. KEGG signaling pathway enrichment analysis resulted in 94 signaling pathways, mainly involving TNF signaling pathway, HIF-1 signaling pathway,Toll-like receptor signaling pathway and FoxO signaling pathway. Conclusion:The mechanism of Zhenwu Decoction in the treatment diabetic kidney disease involves multiple components, targets and pathways, aiming to give Zhenwu Decoction clinical application and deeper research on diabetic nephropathy with a certain theoretical basis.
