      目的:探讨补中益气汤治疗压力性尿失禁(Stress Urinary Incontinence,SUI)的分子作用机制。方法:运用中药系统药理学数据库与分析平台(TCMSP)检索出补中益气汤的药物有效活性成分及其作用靶点。利用GeneGards、OMIM疾病数据库筛选出该病靶点,借助Cytoscape3.7.2软件构建出补中益气汤中药的活性成分-靶点网络和蛋白质-蛋白质相互作用(PPI)网络,采用R软件对靶基因进行基因本体论(Gene Ontology,GO)分析和京都基因与基因组百科全书(Kyoto Encyclopedia of Genes and Genomes,KEGG)通路的分析。结果:共得到补中益气汤有效活性成分182个,通过116个潜在靶标作用于SUI,富集得到GO中2 115个生物过程条目和144条KEGG通路。结论:补中益气汤主要通过槲皮素、山萘酚、7-甲氧基-2-甲基异黄酮、7-O-甲基异丁香酚、利可考酮、β-谷固醇、福莫宁、美迪紫檀素、柚皮苷、糖醇等活性成分调节SELE、ICAM1、MMP3、MAPK14、CD40LG、PLAU、CCNA2、ERBB3、CASP1关键靶点,调控糖尿病并发症中的AGE-RAGE信号通路、膀胱癌信号通路、流体剪切应力与动脉粥样硬化信号通路、TNF信号通路、前列腺癌信号通路、胰腺癌信号通路、乙型肝炎信号通路等多条通路来治疗SUI。
English Summary:
      To explore the molecular mechanism of Buzhong Yiqi Decoction in the treatment of stress urinary incontinence(SUI).Methods:Effective active ingredients and action targets of Buzhong Yiqi Decoction were retrieved with the aid of TCMSP while the disease targets were screened out using GeneGards and OMIM databases.The active ingredient of Buzhong Yiqi Decoction-target network and PPI network were established using Cytoscape 3.7.2 software.And then using R software the gene function of the target gene(Gene Ontology,GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis were annotated.Results:A total of 182 effective active ingredients of Buzhong Yiqi Decoction were obtained,and 116 potential targets can act on SUI.GO functional enrichment analysis obtained 2 115 GO itemsin total.KEGG pathway enrichment analysis obtained 20 pathways.Conclusion:Buzhong Yiqi Decoction mainly through quercetin,kaempferol,7-methoxy-2-methyl isoflavone,7-O-methyl isoeugenol,riccocodone,β-sitosterol,formonine,medicinal pterostilbene,naringin,sugar alcohol and other active ingredients regulates key targets of SELE,ICAM1,MMP3,MAPK14,CD40LG,PLAU,CCNA2,ERBB3,CASP1,and regulates AGE-RAGE in diabetic complications Signal pathways,bladder cancer signaling pathways,fluid shear stress and atherosclerosis signaling pathways,TNF signaling pathways,prostate cancer signaling pathways,pancreatic cancer signaling pathways,hepatitis B signaling pathways and other pathways so that SUI can be treated.
