      目的:基于网络药理学探讨中药复方定眩汤治疗颈性眩晕的分子机制。方法:通过中药系统药理学数据库与分析平台(TCMSP)数据库和BATMAN-TCM数据库筛选药物相关靶点,通过国际公认疾病基因数据库筛选疾病相关靶点,取交集后获得直接靶点并在STRING数据库中进行扩充,后进行网络拓扑分析获得定眩汤治疗颈性眩晕的关键靶点,进行基因本体(GO)富集分析和京都基因和基因组百科全书(KEGG)富集分析,以进一步分析其治疗颈性眩晕的分子机制。结果:在TSMSP数据库与BATMAN-TCM数据库中共提取出活性成分所对应的靶点350个。在疾病基因数据库中得到与颈性眩晕相关的已知靶点253个。2组靶点映射后得到定眩汤作用于颈性眩晕的直接靶点21个,经STRING数据库扩充后得到199个直接和间接靶点。在Cytoscape中进行网络拓扑分析共筛选出99个关键节点,GO分析结果表明,关键节点的生物过程主要包括细胞增殖、细胞凋亡等;细胞成分主要包括核染色质、细胞外空间等;分子功能主要包括酶结合、DNA结合等。涉及的信号通路有MAPK信号通路、PI3K-Akt信号通路、VEGF信号通路等。结论:定眩汤治疗颈性眩晕具有多成分、多靶点的特点;研究初步探讨了定眩汤作用的关键靶点及涉及的生物学过程和信号通路,发现其主要作用可能是影响骨平衡、调节骨代谢、促进血管的修复、调控相关炎症反应。
English Summary:
      To explore the possible molecular mechanism of Dingxuan Decoction in the treatment of cervical vertigo based on network pharmacology.Methods:Drug-related targets through the TCMSP database and BATMAN-TCM database were screened,disease-related targets through the internationally recognized disease gene database were screened,direct targets were obtained after the intersection and expanded them in the STRING database,and then network topology analysis was performed to obtain the key target of Dingxuan Decoction for the treatment of cervical vertigo.GO and KEGG function enrichment analysis were carried out to further analyze the molecular mechanism of the treatment of cervical vertigo.Results:A total of 350 targets corresponding to the active ingredients were extracted from the TSMSP database and the BATMAN-TCM database.In the disease gene database,253 known targets related to cervical vertigo were obtained.After the 2 sets of targets were mapped,21 direct targets of Dingxuan Decoction for cervical vertigo were obtained.After the STRING database was expanded,199 direct and indirect targets were obtained.A total of 99 key nodes were screened out by network topology analysis in Cytoscape,and GO analysis was performed.The results showed that the biological processes of key nodes mainly include cell proliferation,apoptosis,etc.cell components mainly include nuclear chromatin,extracellular space,etc.The molecular functions mainly include enzyme binding,DNA binding and so on.The signal pathways involved were:MAPK signal pathway,PI3K-Akt signal pathway,VEGF signal pathway,etc.Conclusion:The treatment of cervical vertigo with Dingxuan Decoction has the characteristics of multiple components and multiple targets.The key targets,biological processes and signaling pathways involved in the action of Dingxuan decoction are preliminarily investigated,and it is found that its main functions might be to affect bone balance,regulate bone metabolism,promote vascular repair and regulate the process of related inflammation.
