| 目的：研究养元通络针法对糖尿病胃轻瘫（DGP）模型大鼠胃窦平滑肌SCF-kit通路系统及血清胃促生长素(Ghrelin)、胆囊收缩素(CCK)、5-羟色胺表达的影响。方法：将50只SD大鼠随机分为空白组（N）、模型组（M）、针刺组（A）、假针刺组（SA）、西药二甲双胍组（MET），每组10只。除空白组外，其余4组均采用链脲佐菌（STZ）结合高脂饮食诱导造模。各组大鼠分别于相应干预措施2周后，光镜下观察大鼠胃窦平滑肌形态学改变，酶联免疫吸附试验（ELISA）法检测血清Ghrelin、CCK、5-羟色胺表达，Western Blotting法检测胃窦干细胞因子(SCF)、c-kit蛋白表达。结果：1）形态组织学改变：与空白组比较，模型组胃窦黏膜不完整，血管数量减少，腺体的排列紊乱，间质细胞数目减少；与模型组比较，针刺组与西药组胃窦黏膜尚完整，腺体排列比较整齐，有较丰富血管，间质细胞数量较多；假针刺组与模型组比较，差异无统计学意义（P>0.05）。2）与空白组比较，模型组大鼠血清Ghrelin、5-羟色胺表达下降（P<0.05）,CCK、血糖水平升高（P<0.05）；与模型组比较，针刺组与西药组血清Ghrelin、5-羟色胺表达上升,CCK、血糖水平下降（P<0.05），假针刺组Ghrelin、CCK、5-羟色胺表达与模型组比较，差异无统计学意义（P>0.05）。3)与空白组比较，模型组大鼠胃窦SCF、c-kit蛋白表达降低（P<0.05）；与模型组比较，针刺组与西药组大鼠胃窦SCF、c-kit蛋白表达升高（P<0.05）；假针刺组胃窦SCF、c-kit蛋白表达与模型组比较，差异无统计学意义（P>0.05）。结论:养元通络针法能使DGP大鼠胃窦组织内SCF和c-kit蛋白表达量提升,上调SCF-kit信号系统，提高血清Ghrelin、5-羟色胺表达，降低血清CCK表达，从而改善胃肠动力。 |

| To study the effects of Nourishing Yuan and Dredging the Meridians Acupuncture on the SCF-kit pathway system of gastric antrum smooth muscle and the expression of serum Ghrelin,CCK and 5-HT in diabetic gastroparesis model rats.Methods:A total of 50 SD rats were randomly divided into a blank group(N),a model group(M),an acupuncture group(A),a sham acupuncture group(SA),and a metformin group(MET),with 10 rats in each group.Except the blank group,the other 4 groups were induced by streptozotocin(STZ) and combined with high-fat diet to induce modeling.Rats in each group were observed for 2 weeks after the corresponding intervention measures.Morphological changes of gastric antrum smooth muscle were observed under light microscope.Serum Ghrelin,CCK,5-HT expression was detected by ELISA method,and SCF and c-kit proteins were detected by WB method.Results:1) Morphological and histological changes:Compared with the blank group,the gastric antrum mucosa of the model group was incomplete,the number of blood vessels was reduced,the glands were arranged disorderly,and the number of interstitial cells were reduced.Compared with the model group,the gastric antrum mucosa of the acupuncture group and the western medicine group was basically complete,the glands were arranged neatly,there were abundant blood vessels and a large number of interstitial cells; there was no significant difference in the sham acupuncture group.2) Compared with the blank group,the expression of serum Ghrelin and 5-HT in the model group decreased(P<0.05),CCK and the level of blood glucose increased(P<0.05).Compared with the model group,the expression of serum Ghrelin and 5-HT increased,the level of CCK and blood glucose decreased in the acupuncture group and the Western medicine group,but there was no significant difference in the expression of Ghrelin,CCK and 5-HT in the sham acupuncture group(P>0.05).3) Compared with the blank group,the expression of SCF and c-kit protein in the gastric antrum of the model group decreased(P<0.05).Compared with the model group,the expression of SCF and c-kit protein in the gastric antrum of the acupuncture group and the Western medicine group increased(P<0.05),but there was no significant difference in the expression of SCF and c-kit protein in the gastric antrum of the sham acupuncture group(P>0.05).Conclusion:The method of mourishing Yuan and dredging the meridians acupuncture can increase the expression of SCF and c-kit protein in gastric antrum of DGP rats,up-regulate SCF-kit signal system,increase the expression of serum Ghrelin and 5-HT,and decrease the expression of serum CCK,so as to improve gastrointestinal motility. |
