      目的:观察钩藤碱固体脂质纳米粒(Rhy-SLN)对小鼠气道平滑肌细胞(ASMC)的增殖细胞核抗原(PCNA)表达以及TGF-β1/Smad信号通路的影响。方法:将小鼠ASMC体外分离培养,用Rhy-SLN干预ASMC,用免疫荧光法测定小鼠ASMC的PCNA的表达;Western Blotting法检测小鼠ASMC中Smad2、p-Smad2、Smad3、p-Smad3、Smad4、Smad7蛋白表达。结果:Rhy-SLN在一定程度上可以降低小鼠气道平滑肌细胞中PCNA的表达1.80±0.12比1.50±0.15(P<0.05),显著下调Smad2蛋白1.09±0.20、p-Smad2蛋白1.97±0.70、p-Smad3蛋白4.47±0.35、p-Smad4蛋白1.42±0.20相对表达(P<0.05),显著上调Smad7蛋白0.65±0.15相对表达(P<0.05),效果与通路阻滞剂SB431542组相似。结论:Rhy-SLN可以抑制小鼠ASMC的增殖,其机制可能与调节TGF-β1诱导的Smad信号通路有关。
English Summary:
      To observe the effect of Rhy-SLN on the expression of PCNA and TGF-β1/Smad signal pathway of ASMC in mice. Methods:Mouse ASMC were isolated and cultured in vitro, Rhy-SLN interfered with ASMC, and the expression of PCNA in mouse ASMC was determined by immunofluorescence method; the expression of Smad2, p-Smad2, Smad3, p-smad3, Smad4, Smad7 in ASMC was detected by Western blot. Results:To a certain extent, Rhy-SLN can reduce the expression of PCNA in airway smooth muscle cells of mice [(1.80±0.12) VS (1.50±0.15)] (P<0.05), significantly down regulate the relative expression of Smad2 (1.09±0.20), p-Smad2(1.97±0.70), p-Smad3(4.47±0.35), p-Smad4(1.42±0.20) protein (P<0.05), significantly up regulate the relative expression of Smad7 (0.65±0.15) protein (P<0.05), the effect was similar to that of SB431542. Conclusion:Rhy-SLN can inhibit the proliferation of ASMC in mice, and its mechanism may be related to the regulation of Smad signal pathway induced by TGF-β1.
