      目的:通过网络药理学方法探讨小青龙汤治疗变应性鼻炎(Allergic Rhinitis,AR)的作用机制。方法:运用中药系统药理学数据库与分析平台(TCMSP)获得小青龙汤的活性成分及作用靶点,在疾病靶点数据库GeneCards、OMIM获得变应性鼻炎的靶点集,通过R语言将其与中药靶点相映射,从中筛选出共同活性靶点,运用Cytoscape软件、BisoGenet插件构建中药-活性成分-靶点多维网络及小青龙汤-变应性鼻炎蛋白质-蛋白质相互作用(PPI)网络,运用CytoNCA插件、R语言分别进行网络拓扑分析、进行基因本体(GO)富集分析和京都基因和基因组百科全书(KEGG)通路富集分析。结果:获得核心药对“麻黄-甘草”,核心活性成分槲皮素、山柰酚、木犀草素、黄芩素、柚皮素等,核心作用靶点NTRK1、MCM2、CUL3、ESR1、TP53等,涉及通路为AGE-RAGE信号通路、KSHV、TNF信号通路、人巨细胞病毒感染、乙肝、IL-17信号通路、Th17细胞分化等。结论:小青龙汤可通过干预AGE-RAGE等信号通路抑制核因子κB核基因表调节基因转录,通过介导IL-17细胞因子调节Th17细胞分化进一步调节Th1/Th2平衡达到抗炎、调节免疫作用。
English Summary:
      To discuss the mechanism of Xiaoqinglong Decoction (XQL Decoction) in the treatment of allergic rhinitis by network pharmacology.Methods:The ingredients targets of XQL Decoction were acquired from Traditional Chinese Medicine Systems Pharmacology Databaseand Analysis Platform(TCMSP),AR targets were obtained from GeneCards and OMIM database.Then the AR targets and ingredients targets were mutual mapped to find the commontargets,Cytoscape(3.7.1) was used to establish herb-ingredients-targets network and protein-protein interaction network(ppi) and network topology analysis R version was used to gain the GO and KEGG enrichment analysis.Results:Ephedra-liquorice is core herb pair; core ingredients are quercetin,kaempferol,luteolin,baicalein,naringenin,etc.Core targets are NTRK1,MCM2,CUL3,ESR1,TP53,UBC,etc.AGE-RAGE,KSHV,TNF signaling pathway,Hu-man cytomegalovirus infection,hepatitis B,IL-17 signaling pathway,Th17 cell differentiation are involved.Conclusion:XQL Decoction can inhibit NF-κB nuclear gene table regulation gene transcription by interfering with AGE-RAGE and other signaling pathways,and regulate Th1/Th2 balance by mediating IL-17 cytokines to regulate Th17 cell differentiation to achieve anti-inflammatory and immune regulation effects.
