      目的:研究抗抑郁症中药新药-参味宁郁片对慢性应激抑郁模型大鼠及小鼠悬尾抑郁模型的抗抑郁作用。方法:大鼠慢性应激模型:除正常组外,其余各组给予相应的慢性应激,并在造模前1 h灌胃给药,共持续21 d。实验前后通过体质量变化、糖水实验、自发活动开场实验测试,观察各组大鼠相应指标的评分变化。采用放射免疫测定法检测相关激素水平。小鼠悬尾抑郁模型:连续给药7 d后用小鼠自主活动记录仪记录活动次数,再将小鼠悬尾后记录小鼠不动时间,探究各给药组对小鼠悬尾不动时间的影响。结果:慢性应激抑郁模型:在造模21 d后,与正常组比较,模型组大鼠在体质量、糖水偏好指数、水平及垂直运动评分等方面均显著降低(P<0.05),参味宁郁片低剂量组在糖水实验、水平及垂直运动评分与模型组比较差异均有统计学意义(均P<0.05)。此外,参味宁郁片3个剂量组均可有效降低慢性应激模型大鼠CRF的含量,低剂量组可显著降低模型大鼠的ACTH水平(P<0.05)。小鼠悬尾抑郁模型:各给药组小鼠悬尾后的不动时间与模型组比较差异均有统计学意义(均P<0.05)。结论:抗抑郁症中药新药-参味宁郁片对慢性应激抑郁模型及小鼠悬尾抑郁模型均具有一定的抗抑郁作用。
English Summary:
      To investigate the antidepressant effect of Shenwei Ningyu Pills,a new anti-depression Chinese medicial,on the chronic stress depression model with rat and mouse tails suspended.Methods:Chronic Stress Model in Rats:except for the normal control group,rats in other groups were given the corresponding chronic stress,and given by gavage 1 h before modeling,for a total of 21 d.The changes of each index before and after the experiment were observed through body weight change,sugar water test and open field test.The levels of related hormones were detected by radioimmunoassay.Mouse suspended tail depression model:After continuous administration for seven days,the activity times were recorded with the mouse automatic activity recorder,and then the mouse fixed time was recorded after tail suspension,to explore the effects of each administration group on the suspended tail fixed time of mice.Results:Chronic stress depression model:Twenty-one days after modeling,compared with the normal group,rats in the model group exhibited significantly reduced body weight,sucrose preference index,and horizontal and vertical motor scores (P<0.05).Compared with the model group,the low-dose Shenwei Ningyu Pills group had significant differences in the sugar water test,horizontal and vertical movement scores (Ps<0.05).In addition,all three dose groups of Shenwei Ningyu Pills could effectively reduce the content of CRF in chronic stress model rats,and the low dose group could significantly reduce the ACTH level in model rats (P<0.05).Mouse suspended tail depression model:The immobility time after tail suspension in each administration group was significantly different from that in the model group (Ps<0.05).Conclusion:Shenwei Ningyu Pills,a new anti-depression Chinese medicinal,has a certain anti-depression effect on both the chronic stress depression model and the mouse suspended tail depression model.
