      目的:基于数据挖掘和网络药理学探究中医古方治疗冠心病的配伍规律及作用机制。方法:从国家人口与健康科学数据共享平台提供的中医古方数据集V1.0中筛选治疗冠心病的方剂,通过SPSS Statistics25.0、SPSS Modeler14.1等软件进行数据挖掘,总结中医古方治疗冠心病的用药规律。利用中药系统药理学数据库与分析平台(TCMSP)、GeneCards等数据库,筛选核心药物的有效成分、作用靶点及冠心病相关靶点,使用Cytoscape 3.2.7软件构建核心药物治疗冠心病“有效活性成分-靶点-疾病”的网络图;通过David数据库进行基因本体(GO)和京都基因和基因组百科全书(KEGG)富集分析。结果:筛选得到141首方剂,总计313味中药,其中排名前10的药物为甘草、人参、当归、茯苓、肉桂、白芍、白术、陈皮、川芎、干姜;因子分析得到8个公因子,通过关联规则分析得到核心药物“当归-白芍-川芎”。对核心药物进行网络药理学分析得到24个有效活性成分和5个核心作用靶点;KEGG富集通路主要富集在MAPK、Toll样受体、PI3K-AKT、TNF等信号通路。结论:古代医家治疗冠心病多采用补虚药、理气药、活血化瘀药,并主要以补益药为主,其核心药物主要通过调节AKT1、MAPK1等基因及PI3K-AKT信号通路、Toll样受体信号通路的表达实现治疗冠心病的目的。
English Summary:
      To explore the compatibility and mechanism of traditional Chinese medicine in the treatment of coronary heart disease based on the methods of data mining and network pharmacology.Methods:Prescriptions for coronary heart disease were screened from the data set v1.0 provided by the national population and health science data sharing platform.Data mining was carried out through software such as SPSS Statistics 25.0,SPSS Modeler 14.1,etc.,and the rule of medication of traditional Chinese medicine(TCM) for the treatment of coronary heart disease was summarized.TCMSP,GeneCards and other databases were used to screen the active ingredients,targets of action and coronary heart disease-related targets of core drugs,and Cytoscape 3.2.7 software was used to construct a network diagram of “effective active ingredients-targets-disease” of core drugs for the treatment of coronary heart disease; David database was used for GO and KEGG enrichment analysis; Results:A total of141 prescriptions were screened,and 313 Chinese medicinals were obtained.The top 10 drugs were Radix et Rhizoma Glycyrrhizae,Radix et Rhizoma Ginseng,Radix Angelicae Sinensis,Poria,Cortex Cinnamomi,Radix Paeoniae Alba,Rhizoma Atractylodis Macrocephalae,Pericarpium Citri Reticulatae,Rhizoma Chuanxiong,Rhizoma Zingiberis.A total of 8 common factors were obtained by factor analysis,the core drug “Angelica sinensis-Radix Paeoniae Alba-Ligusticum wallichii” was obtained by association rule analysis.A total of 24 active components and 5 core targets were obtained by network pharmacology analysis of core drugs.KEGG enrichment pathway was mainly in MAPK signaling pathway,Toll-like receptor signaling pathway,PI3K-AKT signaling pathway,TNF signaling pathway etc.Conclusion:In ancient times,ancient physicians treats coronary heart disease by Chinese medicinal of tonifying deficiency,regulating qi and promoting blood circulation to remove blood stasis.The core drugs mainly regulate the expression of AKT1,mapk1,PI3K-AKT signaling pathway and Toll-like receptor signaling pathway to treat coronary heart disease.
