目的:研究芦荟粉、火麻仁油及其配伍对便秘小鼠胃肠动力和水液代谢的影响。方法:将160只雄性昆明小鼠随机分为空白组、模型组、果导片组、芦荟粉组(0.15 g/kg)、火麻仁油组(0.33 g/kg)、配伍低剂量组(芦荟粉0.075 g/kg+火麻仁油0.165 g/kg)、配伍中剂量组(芦荟粉0.15 g/kg+火麻仁油0.33 g/kg)、配伍高剂量组(芦荟粉0.3 g/kg+火麻仁油0.66 g/kg)。给药14 d,灌胃复方地芬诺酯5 mg/kg体质量制备小鼠肠蠕动抑制模型。计算各组小鼠小肠墨汁推进率,检测血清5-羟色胺(5-HT)、P物质及血管活性肠肽(VIP)水平变化,采用苏木精-伊红(HE)染色观察结肠组织病理学变化。结果:与空白组比较,模型组小鼠的小肠墨汁推进率显著降低,血清5-HT、P物质水平显著降低,血清VIP水平显著升高,结肠黏膜层上皮细胞和杯状细胞萎缩,黏膜层厚度显著减少,差异有统计学意义(P<0.05);与模型组比较,各给药组小鼠的小肠推进率显著升高(P<0.05),果导片组、芦荟粉组、配伍低剂量组、配伍中剂量组、配伍高剂量组小鼠血清5-HT水平显著升高(P<0.05),果导片组、芦荟粉组、火麻仁油组、配伍中剂量组、配伍高剂量组小鼠血清P物质水平显著升高(P<0.05),果导片组、芦荟粉组、配伍低剂量组、配伍中剂量组、配伍高剂量组小鼠血清VIP水平显著降低(P<0.05),各给药小鼠结肠黏膜层上皮细胞、杯状细胞增加,果导片组、配伍中剂量组、配伍高剂量组小鼠结肠黏膜层厚度显著增加(P<0.05)。结论:芦荟粉、火麻仁油及二者配伍能改善小鼠便秘症状,可通过调节血清5-HT、P物质、VIP等神经递质的分泌,增加肠道动力和调节水液代谢而发挥通便作用。 |

To study the effects of Aloe powder,Cannabis Fructus oil,and their compatibility on gastrointestinal mobility and water metabolism in mice with constipation.Methods:A total of 160 male Kunming mice were randomly divided into a blank group,a model group,a phenolphthalein tablet group,an Aloe powder group(0.15 g/kg),a Cannabis Fructus oil group(0.33 g/kg),a low-dose compatibility group(0.075 g/kg Aloe powder+0.165 g/kg Cannabis Fructus oil),a medium-dose compatibility group(0.15 g/kg Aloe powder+0.33 g/kg Cannabis Fructus oil),and a high-dose compatibility group(0.3 g/kg Aloe powder+0.66 g/kg Cannabis Fructus oil).After 14 days of administration,the mouse model of intestinal peristalsis inhibition was established by intragastric administration of compound diphenoxylate tablets.The ink propelling rate in the small intestine of mice was calculated,and the levels of 5-hydroxytryptamine(5-HT),substance P(SP),and vasoactive intestinal peptide(VIP) in the serum were measured.The pathological changes in colon tissues were observed by hematoxylin-eosin(HE) staining.Results:Compared with the blank group,the model group had a significantly lower intestinal ink propulsion rate,lower 5-HT and SP levels,and a significantly higher VIP level in the serum.The atrophy of colonic mucosal epithelial cells and goblet cells was observed in the model group,and the mucosal thickness was significantly reduced(P<0.05).As compared with the model group,the small intestine ink propulsion rate of each treatment group was significantly increased(P<0.05).The level of 5-HT in the serum of the phenolphthalein tablet group,Aloe powder group,and low,medium,and high-dose compatibility groups was significantly increased(P<0.05).The level of SP in the serum of the phenolphthalein tablet group,Aloe powder group,Cannabis Fructus oil group,and medium and high-dose compatibility groups was significantly increased(P<0.001,P<0.01,P<0.05); the contents of VIP in serum of phenolphthalein tablets group,aloe vera leaf powder group and low,medium and high compatibility groups was significantly decreased(P<0.05).The level of VIP in the serum of the phenolphthalein tablet group,Aloe powder group,Cannabis Fructus oil group,and low,medium,and high-dose compatibility groups was significantly decreased(P<0.05).The epithelial cells and goblet cells of colon mucosa layer of mice in treatment groups were increased,and the thickness of colonic mucosa in the phenolphthalein tablet group and medium and high-dose compatibility groups was significantly increased(P<0.05).Conclusion:Aloe powder,Cannabis Fructus oil,and their compatibility can improve the symptoms of constipation,which may be related to the regulation of SP,5-HT,VIP,and other neurotransmitters in the serum,the regulation of water metabolism,and the increase of intestinal motility. |
