目的:探讨电针“梨状二穴”对腰椎间盘突出症大鼠血清白细胞介素-6(IL-6)、一氧化氮(NO)、前列腺素E2(PGE2)、基质金属蛋白酶-3(MMP-3)等炎症介质表达的影响。方法:将32只SD大鼠随机分为空白组、模型组、电针1组、电针2组。除空白组外,其余3组均按照非压迫性腰椎间盘髓核突出模型的造模方法进行造模。电针1组、电针2组分别给予相应干预措施,在造模结束以及治疗后的第1、3、6天对大鼠后肢热刺激缩爪阈差值进行检测,治疗后的第6天用酶联免疫吸附试验(ELISA)测定血清标本中IL-6、NO、PGE2、MMP-3的水平。结果:造模后第3天,与空白组比较,模型组、电针1组、电针2组大鼠的缩爪阈值明显下降(P<0.01);治疗后第6天,模型组治疗前后缩爪阈值比较,差异无统计学意义(P>0.05);电针1组、电针2组治疗前后比较,缩爪阈值明显升高,差异均有统计学意义(均P<0.01)。治疗后电针1组、电针2组IL-6、NO、PGE2、MMP-3水平明显低于模型组,差异均有统计学意义(均P<0.01)。结论:电针“梨状二穴”降低了腰椎间盘突出症大鼠IL-6、NO、PGE2、MMP-3的表达水平,从而起到抗炎镇痛的作用。 |

To investigate the effects of electroacupuncture at two points of piriform muscle on the expression of interleukin-6(IL-6),nitric oxide(NO),prostaglandin 2(PGE2),and matrix metalloproteinase 3(MMP-3) in the serum of model rats of lumbar disc herniation.Methods:A total of 32 SD rats were randomly assigned into a blank group,a model group,and electroacupuncture groups 1 and 2,with 8 rats in each group.Except the blank group,the other 3 groups were modeled according to the modeling method of non-compressive model of nucleus pulposus of lumbar intervertebral disc.Electroacupuncture groups 1 and 2 were treated with the corresponding intervention measures.At the end of modeling and on the 1st,3rd,and 6th days of treatment,the hind paw withdrawal threshold was measured.The serum levels of IL-6,NO,PGE2,and MMP-3 were measured by enzyme-linked immunosorbent assay(ELISA) after 6 days of treatment.Results:Three days after modeling,the paw withdrawal thresholds in the model group and electroacupuncture groups 1 and 2 declined compared with that in the blank group(P<0.01).The paw withdrawal threshold in the model group showed no significant difference before and after 6 days of treatment(P>0.05).The electroacupuncture increased the paw withdrawal threshold compared with that before treatment(P<0.01).The levels of IL-6,NO,PGE2,and MMP-3 in electroacupuncture groups 1 and 2 were lower than those in the model group(P<0.01).Conclusion:Electroacupuncture at two points of piriform muscle can down-regulate the expression of IL-6,NO,PGE2,and MMP-3 in the serum to exert the anti-inflammatory and pain-relieving effects on lumbar disc herniation in model rats. |
