目的:研究荆防颗粒对脂多糖(LPS)致小鼠急性肺损伤(ALI)模型的预防和保护作用,并探讨其作用机制。方法:将雄性C57BL/6小鼠随机分为对照组、模型组、荆防颗粒低剂量组(1.25 g/kg)、荆防颗粒中剂量组(2.5 g/kg)、荆防颗粒高剂量组(5 g/kg),每组10只。各组分别给予同体积生理盐水和相应浓度荆防颗粒灌胃。给药6 d后,模型组和各给药组采用气管滴注LPS的方法建立ALI模型,对照组给予气管滴入相同体积生理盐水。分析各组肺组织宏观形态学变化;采用苏木精-伊红(HE)染色法观察肺组织病理变化并进行病理学评分;检测肺组织单核细胞趋化蛋白-1(MCP-1)、肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)等炎症指标的变化;进一步检测肺组织髓过氧化物酶(MPO)、诱导型一氧化氮合酶(iNOS)、丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽(GSH)等氧化应激指标的变化。结果:与对照组比较,模型组小鼠肺组织中MCP-1、TNF-α、IL-1β、IL-6、MPO、iNOS和MDA的含量显著升高(P<0.01),SOD和GSH活性水平下降(P<0.01),肺组织病理学评分显著升高(P<0.01),宏观形态受损严重;与模型组比较,荆防颗粒高、中、低剂量组均能不同程度降低肺组织中MCP-1、TNF-α、IL-1β、IL-6、MPO、iNOS和MDA的含量(P<0.05),提高SOD和GSH的活性水平(P<0.05),肺组织病理学评分显著下降(P<0.01),宏观形态得到有效改善。结论:荆防颗粒可在一定程度上抑制LPS所诱导的ALI,改善肺组织宏观形态以及病理学变化,其作用机制可能与抑制炎症介质的分泌、降低氧化应激水平以及提高机体的抗氧化能力有关。 |

To study the preventive and protective effects of Jingfang Granules on lipopolysaccharide(LPS)-induced acute lung injury in mice,and to preliminarily explore the mechanism.Methods:Male C57BL/6 mice were randomly divided into control group,model group,and Jingfang Granules low-,medium-,and high-dose groups(1.25 g/kg,2.5 g/kg,and 5 g/kg,respectively),10 in each group.Mice in each group were separately given normal saline of the same volume and Jingfang Granules of corresponding concentrations.After 6 d of administration,the model of acute lung injury was established by endotracheal infusion of LPS in model group and Jingfang Granules groups,while the control group received the same volume of normal saline by endotracheal infusion.The macroscopic morphological changes of lung tissues in each group were analyzed.HE staining was used to observe the pathological changes of lung tissues and the pathological score was given.Monocyte chemotactic protein 1(MCP-1),interleukin-1β(IL-1β),interleukin-6(IL-6) and tumor necrosis factor-α(TNF-α) in lung tissues were detected.Oxidative stress indicators such as myeloperoxidase(MPO),inducible nitric oxide synthase(iNOS),malondialdehyde(MDA),superoxide dismutase(SOD),and glutathione(GSH) were also detected.Results:Compared with the the conditions in control group,MCP-1,TNF-α,IL-1β,IL-6,MPO,iNOS and MDA in the lung tissues of mice in model group as well as the pathological score were increased(P<0.01),with severe macroscopic morphological changes,and the activities of SOD and GSH were decreased(P<0.01).Compared with model group,Jingfang Granules groups had down-regulated MCP-1,TNF-α,IL-1β,IL-6,MPO,iNOS and MDA to different degrees(P<0.05),and increased activities of SOD and GSH(P<0.05).The pathological score of lung tissues was lowered(P<0.01),and the macroscopic morphology was improved.Conclusion:Jingfang Granules can inhibit LPS-induced acute lung injury to a certain extent and improve the macroscopic morphology and pathological changes of lung tissues.The mechanism may be related to inhibiting the inflammatory factors,reducing the oxidative stress,and enhancing the antioxidant capacity of the body. |
