目的:探讨芍甘附子汤加味对类风湿关节炎骨骼肌损害的作用机制。方法:将36只无特定病原体级(SPF)级雌性Wistar大鼠随机分为正常组、模型组、对照组及中药低、中、高剂量组,每组6只。除正常组,其余各组复制胶原诱导性关节炎(CIA)寒证模型,给相应药物灌胃。灌胃结束取大鼠血清用酶联免疫吸附试验法(ELISA)检测肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、白细胞介素-1β(IL-1β、白细胞介素-15(IL-15)含量,取腓肠肌,测定湿重、腓肠肌肌环指蛋白1(MuRF1)、肌肉萎缩F盒蛋白(MAFbx)表达。结果:模型组大鼠腓肠肌湿重、质量指数、纤维横截面积较正常组显著下降,加味芍甘附子汤中、高剂量组腓肠肌湿重、质量指数、纤维横截面积较模型组明显增加;且中药高剂量组腓肠肌纤维横截面积与对照组比较差异有统计学意义(P<0.05)。与正常组比较,模型组大鼠血清TNF-α、IL-6、IL-1β、IL-15含量显著升高,腓肠肌MAFbx、MuRF1表达显著升高(P<0.05);与模型组比较,中药高剂量组及对照组TNF-α、IL-6、IL-1β、IL-15,腓肠肌MAFbx、MuRF1表达明显下降;中药组与对照组比较差异无统计学意义(P>0.05)。结论:芍甘附子汤加味可减轻类风湿关节炎骨骼肌损害,其可能作用机制是减少TNF-α、IL-6、IL-15、IL-1β炎症介质的含量和降低MuRF1、MAFbx表达。 |

To explore the mechanism of modified Shaogan Fuzi Decoction on skeletal muscle damage in rheumatoid arthritis.Methods:A total of 36 specific pathogen-free(SPF) female Wistar rats were randomly divided into a normal group,model group,control group,and low,medium,and high dose groups of traditional Chinese medicine(TCM),with six rats in each group.Except for the normal group,other groups were used to replicate the cold syndrome model of collagen-induced arthritis(CIA) and administered with the corresponding drugs.At the end of the administration,rat serum was collected,and the content of tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),interleukin-1β(IL-1β),and interleukin-15(IL-15) was detected by enzyme-linked immunosorbent assay(ELISA).The gastrocnemius muscle was taken,and the wet weight,gastrocnemius muscle-specific ring finger protein1(MuRF1),and muscle dystrophinF-box(MAFbx) expression were measured.Results:The wet weight,mass index,and fiber cross-sectional area of the gastrocnemius of muscle rats in the model group were significantly lower than those in the normal group,and the wet weight,mass index,and fiber cross-sectional area of the gastrocnemius muscle of rats in the medium and high dose groups of modified Shaogan Fuzi Decoction were significantly increased compared with those in the model group.In addition,the fiber cross-sectional area of the gastrocnemius muscle in the high dose group of TCM was significantly different from that in the control group(P<0.05).Compared with that in the normal group,the serum content of TNF-α,IL-6,IL-1β,and IL-15 of the rats in the model group was significantly increased,and the expressions of MAFbx and MuRF1 in the gastrocnemius muscle were significantly increased(P<0.05).Compared with that in the model group,the serum content of TNF-α,IL-6,IL-1β,and IL-15 of the rats in the high dose group of TCM and the control group,as well as the expressions of MAFbx and MuRF1 in the gastrocnemius muscle was decreased significantly.There was no statistically significant difference between the TCM group and the control group(P>0.05).Conclusion:Modified Shaogan Fuzi Decoction can reduce skeletal muscle damage in rheumatoid arthritis,and its possible mechanism is to reduce the content of TNF-α,IL-6,IL-15,and IL-1β inflammatory factors and lower the expression of MuRF1 and MAFbx. |
