      目的:探讨南方红豆杉果乙酸乙酯部位对抑郁症模型大鼠的影响及可能作用机制。方法:采用随机数字法将20只雌性斯泼累格·多雷(SD)大鼠随机分为对照组、模型组、红豆杉果组和氟西汀组,每组5只。除对照组外,其余各组采用慢性不可预知温和应激(CUMS)法4周制备抑郁症模型。造模后红豆杉果组给予南方红豆杉果乙酸乙酯提取物200 mg/(kg·d)灌胃,对照组和模型组给予等体积的生理盐水灌胃,共给药4周。在造模结束后对对照组和模型组进行糖水实验和旷场实验,给药结束后对各组大鼠进行糖水实验和旷场实验,检测各组大鼠血清中白细胞介素-1β(IL-1β)、IL-6、IL-18的含量。结果:在造模结束后与对照组比较,模型组大鼠的糖水消耗率明显减少(P<0.01),活动距离明显降低(P<0.01)。实验结束后,与模型组比较,红豆杉果组和氟西汀组大鼠的糖水消耗率明显增加(P<0.01),活动距离明显增加(P<0.01);与对照组比较,模型组血清中IL-1β、IL-6、IL-18的含量均明显升高(P<0.05,P<0.01);与模型组比较,红豆杉果组和氟西汀组血清中IL-1β、IL-6、IL-18的含量均明显下降(P<0.05,P<0.01)。结论:南方红豆杉果乙酸乙酯部位有抗抑郁的作用,同时有抗炎的作用,可能通过降低抑郁症大鼠炎症反应起到缓解大鼠抑郁样行为。
English Summary:
      To investigate the effects of ethyl acetate extract of Taxus chinensis fruit on the depression model of rats and its possible mechanism.Methods:Twenty female SD rats were randomly divided into the normal group,model group,Taxus chinensis fruit group,and fluoxetine group,with five rats in each group.Except for the normal group,the chronic unpredictable mild stress(CUMS) method was used to prepare the depression model in other groups for four weeks.After modeling,the Taxus chinensis fruit group was given 200 mg/(kg·d) of ethyl acetate extract of Taxus chinensis fruit,and the normal group and the model group were given an equal volume of normal saline by intragastric administration for four weeks.After the modeling,the normal group and the model group were subjected to the sugar water experiment and the field test.After the administration,the rats in each group were subjected to the sugar water experiment and the field test,and the serum content of interleukin 1β(IL-1β),IL-6,and IL-18 was detected.Results:Compared with that in the normal group after the modeling,the sugar water consumption of rats in the model group was significantly decreased(P<0.01),and the activity was significantly decreased(P<0.01).After the experiment,compared with those in the model group,the sugar water consumption and activity of rats in the Taxus chinensis fruit group and the fluoxetine group were significantly increased(P<0.01).Compared with that in the normal group,the serum content of IL-1β,IL-6,and IL-18 in the model group was significantly increased(P<0.05,P<0.01).Compared with that in the model group,the serum content of IL-1β,IL-6,and IL-18 in the Taxus chinensis fruit group and the fluoxetine group was significantly decreased(P<0.05,P<0.01).Conclusion:The ethyl acetate extract of Taxus chinensis fruit has both antidepressant and anti-inflammatory effects,which may alleviate depression-like behavior in rats by reducing inflammatory response.
