To observe the effects of Zhonghua Dieda Pills on in vitro thrombosis formation, blood viscosity, platelet aggregation rate in acute blood stasis model rats, and on ear microcirculation in microcirculation disorder model mice to evaluate the blood-activating and stasis-resolving effects of Zhonghua Dieda Pills. Methods:An acute blood stasis model in rats was established by injecting adrenaline and cold water stimulation, and in vitro thrombosis formation and blood viscosity were measured.Platelet aggregation rate in rats was measured using the Born turbidimetric method with an inducer.Microcirculation disorder was induced in mice by tail vein injection of 15% high-molecular dextran, and changes in blood flow velocity and vessel diameter of arterioles and venules in the mouse ear were observed over time. Results:Zhonghua Dieda Pills significantly reduced blood viscosity in blood stasis model rats, with significant reductions in whole blood viscosity at shear rates of 150 S-1,60 S-1,and 5 S-1(P<0.05).Zhonghua Dieda Pills significantly shortened thrombus length and reduced both wet and dry weights of thrombi in blood stasis model rats(P<0.05,P<0.01).Zhonghua Dieda Pills inhibited platelet aggregation, with all doses significantly reducing ADP-induced platelet aggregation rates in rats(P<0.05,P<0.01).Additionally,Zhonghua Dieda Pills significantly improved microcirculation disorder induced by high-molecular dextran in mice, increased blood flow velocity and expanded arteriole and venule diameters at 10,20,and 30 min post-modeling(P<0.05,P<0.01),and reduced red blood cell aggregation(P<0.05,P<0.01).Conclusion:Zhonghua Dieda Pills can significantly improve blood viscosity, inhibit platelet aggregation, and ameliorate microcirculation disorders, demonstrating a good effect in activating blood circulation and resolving blood stasis. |
