To observe the effects of the Bushen Yunpi Formula on the growth of short stature rats and explore its mechanisms of action.Methods:Thirty female and fifteen male rats were housed together overnight.After conception,female rats were randomly assigned to a low-protein diet group and a standard diet group based on a random number table.All pregnant rats were fed a standard diet after delivery.Offspring were weaned at 3 weeks old after removing female pups at 2 weeks old.From the offspring of rats fed a standard diet,10 male pups were randomly selected by a random number table and marked as the normal group.Fifty short stature model pups from the offspring of rats fed a standard diet were divided into the model group,Western medicine group,low-dose group,medium-dose group,and high-dose group,with 10 rats per group.The normal group and model group were given sterile physiological saline by gavage at 1 mL/100 g.The low,medium,and high-dose groups were given Bushen Yunpi Formula at 0.76 g/mL,1.52 g/mL,and 3.04 g/mL by gavage at 1 mL/100 g.The Western medicine group was injected subcutaneously in the abdomen with recombinant human growth hormone injection(0.84 IU/kg).Administration was given once daily at 8:00 a.m.for 3 weeks.Comparisons were made of body length,body mass,bone mineral density(BMD),bone mineral content(BMC),serum procollagen type Ⅰ carboxy-terminal peptide(PICP),serum type Ⅰ collagen cross-linked peptide(ICTP),growth hormone(GH),and insulin-like growth factor-1(IGF-1) among the groups.Results:After treatment,the body length and body mass of rats in each group were higher than those before treatment(P<0.05).The BMD of the low-dose and high-dose groups was higher than that of the model group(P<0.05).The serum PICP levels of the normal group,Western medicine group,medium-dose group,and high-dose group were lower than those of the model group(P<0.05).The serum PICP levels of the three Chinese medicine groups were higher than those of the normal group(P<0.05).The serum ICTP levels of the normal group,three Chinese medicine groups,and Western medicine group were lower than those of the model group(P<0.01).The serum ICTP levels of the three Chinese medicine groups were higher than those of the normal group(P<0.05).The serum GH levels of the normal group,three Chinese medicine groups,and Western medicine group were lower than those of the model group(P<0.01).The serum GH levels of the low-dose and medium-dose groups were higher than those of the Western medicine group(P<0.05),and the serum GH levels of the low-dose group were higher than those of the normal group(P<0.05).The serum IGF-1 levels of the low-dose and medium-dose groups were lower than those of the Western medicine group(P<0.05).Conclusion:Bushen Yunpi Formula can promote the growth of short stature rats in terms of body length and body mass,enhance bone metabolism,increase BMD,and its mechanism of action involves promoting the utilization of serum GH and increasing serum IGF-1 levels to exert a growth-promoting effect. |
