目的:探究加味芪黄饮对膜性肾病(MN)大鼠肾功能和肾组织细胞凋亡的影响。方法:通过改良Border制备MN大鼠模型,随机分为5组并连续给药4周:空白组(20只)和模型组(15只)灌胃给予生理盐水5 mL;贝那普利组(18只)灌胃给予盐酸贝那普利10 mg/kg;加味芪黄饮低、高剂量组(各18只)分别灌胃给予加味芪黄饮4 mg/kg、8 mg/kg。各组均分别于造模后第2到6周末检测大鼠24 h尿蛋白。造模后第6周末,采用苏木精-伊红(HE)染色观察肾脏病理改变;采用免疫组织化学法检测肾组织B细胞淋巴瘤2相关X蛋白(Bax)、B细胞淋巴瘤2(Bcl-2)及肾母细胞瘤抑制基因1(WT-1)的表达水平,腹主动脉取血检测血尿素氮(BUN)、血肌酐(Scr)、人血白蛋白(Alb)、三酰甘油(TG)、总胆固醇(TC)。结果:模型组较空白组24 h尿蛋白、BUN、Scr、TG、TC、Bax蛋白表达水平升高,Bcl-2、WT-1蛋白表达水平、Alb降低(P<0.01);各给药组较模型组24 h尿蛋白、BUN、Scr、TG、TC以及Bax蛋白表达水平均降低,Bcl-2、WT-1蛋白表达水平以及Alb均升高(P<0.05,P<0.01);且加味芪黄饮高剂量组较低剂量组变化更显著。结论:加味芪黄饮可能通过降低Bax蛋白表达水平,提高Bcl-2、WT-1蛋白表达水平,影响细胞凋亡,保护肾脏足细胞,从而延缓膜性肾病大鼠的肾功能损伤。 |

To explore the effect of modified Qihuang Decoction on renal function and renal tissue cell apoptosis in rats with membranous nephropathy(MN).Methods:The MN rat model was established by an improved Border method,and rats were randomly divided into five groups and treated for 4 consecutive weeks.The blank group(20 rats) and the model group(15 rats) were administered 5 mL of physiological saline by gavage.The Benazepril group(18 rats) was administered benazepril hydrochloride at 10 mg/kg by gavage.The low-dose and high-dose modified Qihuang Decoction groups(18 rats each) were administered modified Qihuang Decoction at 4 mg/kg and 8 mg/kg by gavage,respectively.Urine protein levels were measured at the end of weeks 2 to 6.At the end of week 6,renal pathological changes were observed by hematoxylin-eosin(HE) staining.The expression levels of Bcl-2-associated X protein(Bax),B-cell lymphoma 2(Bcl-2),and Wilms' tumor suppressor gene 1(WT-1) in renal tissue were detected by immunohistochemistry.Blood samples were collected from the abdominal aorta to measure blood urea nitrogen(BUN),serum creatinine(Scr),human serum albumin(Alb),triglycerides(TG),and total cholesterol(TC).Results:In the model group,24-hour urine protein,BUN,Scr,TG,TC,and Bax protein expression levels were significantly higher,while Bcl-2,WT-1,and Alb expression levels were significantly lower compared with the blank group(P<0.01).In each treatment group,24-hour urine protein,BUN,Scr,TG,TC,and Bax protein expression levels were significantly lower,while Bcl-2,WT-1,and Alb expression levels were significantly higher compared with the model group(P<0.05,P<0.01).Furthermore,the high-dose modified Qihuang Decoction group showed more significant changes than the low-dose group.Conclusion:Modified Qihuang Decoction may protect podocytes in MN rats by reducing Bax protein expression,increasing Bcl-2 and WT-1 protein expression,and influencing cell apoptosis,thereby delaying renal function impairment in MN. |
