      目的:探讨四物汤经雌激素受体(ER)介导磷脂酰肌醇3-激酶(PI3K)/蛋白激酶B(AKT)通路保护卵巢衰老大鼠心肌的作用及机制。方法:选用4周龄雌性F344/DuCrl-大鼠,分为空白对照组、模型组、阳性对照组、四物汤高剂量组、低剂量组。除空白对照组,其余4组连续腹腔注射4-乙烯基环己烯二环氧化合物(VCD)20 d,阴道涂片观测动情周期。造模成功后,阳性对照组和四物汤组分别灌胃戊酸雌二醇(E2)和高剂量、低剂量四物汤21 d。取大鼠心脏,苏木精-伊红(HE)染色观察心肌组织形态学改变;马松(MASSON)染色观察心肌组织纤维化程度;蛋白质免疫印记法检测心肌组织中雌激素受体α亚型(ERα)、雌激素受体β亚型(ERβ)、PI3K、AKT、Bcl2相关X蛋白(Bax)、B淋巴细胞瘤-2基因(Bcl-2)表达水平。结果:模型组心肌纤维排列疏松,分布不均且断裂,胞质不匀,胶原纤维增多;各观察组可见心肌纤维排列相对整齐紧密,胞质更均匀,胶原纤维较少。模型组心肌组织ERα、ERβ、PI3K、AKT、Bcl-2蛋白表达低(P<0.05),Bax蛋白表达高(P<0.01)。与模型组比较,各观察组心肌组织ERα、ERβ、PI3K、AKT、Bcl-2蛋白表达高(P<0.05);Bax蛋白表达低(P<0.05)。结论:四物汤对VCD诱导卵巢衰老大鼠心肌具有保护作用,其机制可能与通过ER介导PI3K/AKT信号通路抑制心肌细胞凋亡有关。
English Summary:
      To investigate the effects and mechanisms of Siwu Decoction in protecting the myocardium of ovarian aging rats through the estrogen receptor(ER)-mediated phosphatidylinositol 3-kinase(PI3K)/protein kinase B(AKT) pathway.Methods:Four-week-old female F344/DuCrl rats were selected and divided into the blank control group,model group,positive control group,high-dose Siwu Decoction group,and low-dose Siwu Decoction group.Except for the blank control group,the other four groups were intraperitoneally injected with 4-vinylcyclohexene dioxide(VCD) for 20 days,and the estrous cycle was monitored by vaginal smears.After successful modeling,the positive control group and Siwu Decoction groups were respectively gavaged with estradiol(E2) or high-dose/low-dose Siwu Decoction for 21 days.The hearts of rats were collected,and hematoxylin-eosin(HE) staining was performed to observe myocardial tissue morphology changes.Masson's trichrome staining was used to observe the degree of myocardial fibrosis.Protein expression levels of estrogen receptor α(ERα),estrogen receptor β(ERβ),PI3K,AKT,B-cell lymphoma 2(Bcl-2),and Bcl2-associated X protein(Bax) were detected by Western blot.Results:In the model group,myocardial fibers were loosely arranged,unevenly distributed,and fractured,with uneven cytoplasm and increased collagen fibers.In the treatment groups,myocardial fibers were relatively neat and tightly arranged,cytoplasm was more uniform,and collagen fibers were less.The expression levels of ERα,ERβ,PI3K,AKT,and Bcl-2 proteins in myocardial tissue were low in the model group(P<0.05),and Bax protein expression was high(P<0.01).Compared with the model group,the expression levels of ERα,ERβ,PI3K,AKT,and Bcl-2 proteins were higher in all treatment groups(P<0.05),while Bax protein expression was lower(P<0.05).Conclusion:Siwu Decoction protects the myocardium of VCD-induced ovarian aging rats,and its mechanism may be related to inhibiting myocardial cell apoptosis through the ER-mediated PI3K/AKT signaling pathway.
