目的:研究藿香正气贴敷神阙穴对寒湿腹泻幼龄大鼠的防治效果和作用机制。方法:选取4周龄无特定病原体(SPF)级斯泼累格·多雷(SD)幼龄大鼠,雌雄各半,寒湿环境结合番泻叶诱导腹泻大鼠模型。将大鼠随机分为空白对照组、模型组、阳性药组、藿香正气水灌胃组、藿香正气贴低剂量组、藿香正气贴高剂量组,每组10只。药物干预2周后,采用酶联免疫吸附试验(ELISA)法检测肿瘤坏死因子α(TNF-α),白细胞介素17(IL-17),白细胞介素1β(IL-1β)水平;以苏木精-伊红(HE)染色法观察结肠病理变化;基于16S核糖体核糖核酸(16S rRNA)测序技术检测各组大鼠肠道菌群的差异。结果:与模型组比较,HXZQ-L、藿香正气贴高剂量组均能改善大鼠粪便性状,IL-1β、IL-17、TNF-α水平降低(均P<0.05),藿香正气贴高剂量组肠道菌群的比值(F/B)升高(P<0.05),乳杆菌属丰度增加(P<0.05)。结论:藿香正气贴神阙穴贴敷可有效防治幼龄大鼠寒湿腹泻,其作用机制可能与抑制炎症反应、修复肠道黏膜损伤、改善肠道菌群环境有关。 |

To investigate the preventive and therapeutic effects and mechanisms of Huoxiang Zhengqi Plaster applied to Shenque(CV 8) in treating cold-dampness type diarrhea in young rats.Methods:Four-week-old specific pathogen-free(SPF) Sprague-Dawley(SD) rats,with equal numbers of males and females,were used to establish a diarrhea model by cold-dampness environment combined with senna leaves.The rats were randomly divided into six groups:blank control group,model group,positive drug group,oral Huoxiang Zhengqi Liquid group,low-dose Huoxiang Zhengqi Plaster group,and high-dose Huoxiang Zhengqi Plaster group,with 10 rats per group.After 2 weeks of drug intervention,tumor necrosis factor α(TNF-α),interleukin 17(IL-17),and interleukin 1β(IL-1β) levels were measured by enzyme-linked immunosorbent assay(ELISA).Colon pathological changes were observed using hematoxylin-eosin(HE) staining.Differences in the gut microbiota among groups were analyzed based on 16S rRNA sequencing.Results:Compared to the model group,both the low-and high-dose Huoxiang Zhengqi Plaster groups improved stool characteristics,reduced IL-1β,IL-17,and TNF-α levels(all P<0.05),and the high-dose Huoxiang Zhengqi Plaster group showed an increased fecal microbiota ratio(F/B)(P<0.05) and higher abundance of Lactobacillus(P<0.05).Conclusion:Application of Huoxiang Zhengqi Plaster to the Shenque(CV 8) effectively prevents and treats cold-dampness type diarrhea in young rats.Its mechanism of action may involve the inhibition of inflammatory response,repair of intestinal mucosal damage,and improvement of the gut microbiota environment. |
