      目的:探讨祛瘀安神法中药对急性心肌梗死合并焦虑大鼠心房利钠肽(ANP)、利尿钠肽A型受体(NPRA)水平的调节作用。方法:根据随机数字法将大鼠分为假手术组、心肌梗死合并焦虑组(复合模型组)及中药组。采用不确定空瓶刺激法结合冠状动脉左前降支结扎,建立心肌梗死合并焦虑大鼠模型。造模结束后,对各组大鼠进行超声心动及行为学检测,采用酶联免疫吸附测定法检测外周血ANP含量,逆转录聚合酶链式反应(PCR)及免疫印迹试验法检测心房ANP及NPRA水平。结果:1)超声心动结果:与复合模型组比较,中药组大鼠左室射血分数(EF)、左室短轴缩短率(FS)值升高(P<0.01),左室收缩末内径(LVIDs)、左室舒张末内径(LVIDd)下降(P<0.01)。2)行为学检测:旷场实验结果显示与复合模型组比较,中药组大鼠水平运动得分有所上升,差异有统计学意义(P<0.05);高架十字迷宫实验结果显示,与复合模型组比较,中药组大鼠更倾向于待在开臂里,差异有统计学意义(P<0.05)。3)外周血ANP含量:与复合模型组比较,中药组大鼠外周血ANP含量明显下降(P<0.01)。4)心房ANP及NPRA水平:与复合模型组比较,中药组大鼠ANP与NPRA表达含量均明显上升,且差异有统计学意义(P<0.05)。结论:祛瘀安神法中药可有效改善急性心肌梗死合并焦虑大鼠心功能及焦虑状态,考虑与其增加大鼠心房ANP、NPRA表达水平相关。
English Summary:
      To investigate the regulation effect of the Quyu Anshen method on atrial natriuretic peptide(ANP) and natriuretic peptide receptor-A(NPRA) levels in rats with acute myocardial infarction(AMI) and anxiety.Methods:Rats were divided into a sham operation group,a myocardial infarction and anxiety group(compound model group),and a traditional Chinese medicine(TCM) group according to the random number method.The rat model of myocardial infarction and anxiety was established by using uncertain empty water bottle stress and the left anterior descending coronary artery ligation.After modeling,ultrasound cardiograms and behavioral tests were performed on rats in each group.ANP content in peripheral blood was detected by enzyme-linked immunosorbent assay,and ANP and NPRA levels were detected by reverse transcription polymerase chain reaction(PCR) and Western Blotting.Results:1) Ultrasound cardiogram results:Compared with those in the compound model group,the values of ejection fraction(EF) and fractional shortening(FS) in the left ventricle of rats in the TCM group were increased(P<0.01),while left ventricular internal dimension in systole(LVIDs) and left ventricular internal diastolic dimension(LVIDd) were decreased(P<0.01).2) Behavioral test:The results of the open field experiment showed that compared with that in the compound model group,the exercise scores of rats in the TCM group were increased,and the difference was statistically significant(P<0.05).The results of the elevated cross maze experiment showed that compared with those in the compound model group,the rats in the TCM group were more likely to stay in the open arm(P<0.05).3) ANP content in peripheral blood:Compared with that in the compound model group,the ANP content in peripheral blood of rats in the TCM group was significantly decreased(P<0.01).4) ANP and NPRA levels:Compared with those in the compound model group,the expression levels of ANP and NPRA in the TCM group were significantly increased(P<0.05).Conclusion:The Quyu Anshen method can effectively improve the cardiac function and anxiety state of rats with AMI and anxiety,and the mechanism may be related to increased expression levels of ANP and NPRA in rats.
