Abstract:To investigate the optimum combination of Disintoxication and chemotherapy in auxiliary treatment phase on colon cancer xenografts in mice, elucidate the potential mechanism of Chinese herbal for adjuvant chemotherapy.Methods:1) Colon cancer cell line CT 26 was injected subcutaneously on 50 BALB/c mice to establish colon cancer xenografts model, they were randomly divided into 5 groups: 10 BALB/C mice in pre intervention group、10 in post intervention group、10 in Herbal only Herbal group、10 in Chemotherapy only group、10 BALB/C mice in model control, the other 10 mice were used as negative control group. 2) Body weight and tumor volume were measured during the treatment in the BALB/c mice from each groups and negative control group. Harvesting blood from different groups, separating serum, weighting tumor weight and calculating anti tumor rate. 3) Detecting content of CEA、sCD44 in serum from different groups by ELISA.Results:1) Tumor inhibition rate: The anti tumor rate was 50% 60% in optimum combination groups, while 40% in chemotherapy group and 20% in Herbal only group. in sequence of Pre intervention group > Post intervention group > Chemotherapy only group > Herbal only group. 2) Change of mice weight without tumor weight: Compared with the weight before the treatment,mice weight increased after treatment in Pre intervention group and Herbal only group. While the result is opposite in chemotherapy group、post intervention group and model control group. 3) Serum level of CEA and sCD44:Compared with model control group, the content of CEA、sCD44 decreased in treatment groups, while with dramatic decline in two therapy combination group than single drug treatment group. Compared with model control group, the content of CEA、sCD44 in pre intervention group、post intervention group and chemotherapy group had statistical significance (P<005).Conclusion:The mechanism of the combination of detoxicating therapy and chemotherapy inhibits tumor perhaps related to decreasing expression of CEA、sCD44.
