增液汤干预干燥综合征模型鼠AQP1/AQP8的表达规律研究

    摘要:

    目的:观察增液汤干预下干燥综合征(Sjogren's Syndrome,SS)模型小鼠肺及结肠组织中水通道蛋白AQP1(aquaporin1)、AQP8(aquaporin8)的表达规律。方法:选用BALB/c小鼠,各组10只,免疫诱导法建立干燥综合征小鼠模型,产生SS特异性临床表现和病理改变,采用免疫组化和Western blot蛋白质印迹法观察增液汤对SS模型小鼠肺及结肠组织中AQP1/AQP8表达的影响。结果:增液汤可以增强肺及结肠组织中AQP1的蛋白表达(P<0.01),对AQP8无影响。结论:AQP1在干燥综合征模型鼠多组织中的表达下调,可能是干燥综合征水液代谢障碍,多脏器损伤的机制之一;增液汤恢复机体阴液,起到“增水”功效可能与上调AQP1蛋白表达有关。

    Abstract:

    To observe the expressions of AQP1 and AQP8 in lung and colon tissue of mice with Sjogren's syndrome. Methods:Fifty BALB/c mice were randomized into five groups,with10 mice in each group. The mice models with Sjogren's syndrome were established by immune inducing method. It was able to produce the specific clinical symptoms and pathological changes of Sjogren's syndrome. The effect of Zengye decoction on the expression of AQP1/AQP8 in lung and colon tissue was observed by immunohistochemistry and Western blot. Results:Zengye decoction can strengthen the AQP1 protein expression in lung and colon tissue (P<0.01),however AQP8 was barely changed. Conclusion:It could be one of the causes of Sjogren's syndrome that AQP1 expression was reduced in multiple organ in the mice with Sjogren’s syndrome. Zengye decoction can increase the body fluid,and the effect may be associated with the increased AQP1 expression.
