    摘要:

    目的:检测正常大鼠体内物质代谢与能量代谢相关酶的含量,考察桑白皮拆分组分对其表达的影响,以期阐释其寒热属性。方法:SD雄性大鼠随机分为正常组(Control)、寒性药黄连组(HL)、黄芩组(HQ)、黄柏组(HB)、热性药附子组(FZ)、干姜组(GJ)、花椒组(HJ),桑白皮30%乙醇洗脱组分组(S-30)、桑白皮-脂肪油组分组(S-Z)。连续给药9d,留取肝脏、心脏等脏器,检测物质代谢相关酶葡萄糖激酶(GCK)、果糖磷酸激酶(PFK-1)、磷酸甘油酸激酶(PGK)、丙酮酸激酶(PK)、丙酮酸脱氢酶(PDH)、乙酰辅酶A(acetyl-CoA)、柠檬酸合酶(CS)、异柠檬酸脱氢酶(ICD)、α-酮戊二酸脱氢酶(α-KGDHC)、延胡索酸酶(FUM)、糖原磷酸化酶(PYGL)、糖原合成酶激酶-3(GSK-3)、脂肪甘油三酯脂酶(ATGL)及能量代谢相关酶细胞色素C还原酶(CCR)、细胞色素C氧化酶(COX)、ATP合酶(ATPs)、腺苷酸激酶(ADK)、Na+-K+ATP酶的表达和ATP、ADP、NAD+、NADH、NAD+/NADH含量,初步探讨桑白皮拆分组分对物质代谢与能量代谢的影响。结果:与正常组相比,HL、HQ组大鼠体重有显著降低(P<0.05或P<0.01),其他组没有统计学意义(P>0.05);HL、HQ、HB组GCK、PFK-1、PGK、PK、ICD、CCR、COX、ATPs、ADK的表达显著降低(P<0.05或P<0.01),acetyl-CoA、α-KGDHC、PYGL、ATGL的表达显著升高(P<0.05或P<0.01),ATP含量虽有所降低但没有统计学意义(P>0.05),ADP、NAD+、NADH、NAD+/NADH含量均没有统计学意义(P>0.05);FZ、GJ、HJ组GCK、PGK、CCR、COX、ATPs、ADK、ICD的表达显著升高(P<0.05或P<0.01),acetyl-CoA、α-KG DHC、PYGL的表达显著降低(P<0.05或P<0.01),ATP含量虽有所升高但没有统计学意义(P>0.05),ADP、NAD+、NADH、NAD+/NADH含量均没有统计学意义(P>0.05);S-30、S-Z组GCK、PFK-1、PGK、PK、PDH、ICD、GSK-3、CCR、COX、ATPs、ADK的表达显著降低(P<0.05或P<0.01),α-KGDHC的表达显著升高(P<0.05),与寒凉性中药HL、HQ、HB具有相同或相似的作用趋势。结论:典型寒、热中药可以影响机体物质能量代谢,且寒性药能够抑制物质能量代谢,热性药能够促进物质能量代谢,这与本课题首席科学家匡海学教授提出的新假说相契合;总体来看,S-30、S-Z组也能够抑制物质能量代谢,与寒性药作用类似,可初步推断其药性可能为寒凉。

    Abstract:

    To explore the cold and warm property of fraction of Mori Cortex by detecting related enzyme activity in substance and energy metabolism in normal rats.Methods:SD rats were randomly divided into normal,cold medicine Coptis chinensis Franch group,Scutellaria baicalensis Georgi group,Phellodendron chinense Schneid.Group,and warm medicine Aconitum carmichaelii Debx group,Zingiber offcinale Rosc group,Zanthoxylum bungeanum Maxim.group,30% split fraction of Mori Cortex group,fatty oil fraction of Mori Cortex group.All groups were administered orally for 9 days and detected glucokinase(GCK),phosphofructokinase(PFK-1),phosphoglycerate kinase(PGK),pyruvate kinase(PK),pyruvate dehydrogenase(PDH),acetyl coenzyme A(acetyl-CoA),citrate synthase(CS),isocitrate dehydrogenase(ICD),α-ketoglutarate dehydrogenase(α-KGDHC),fumarase(FUM),glycogen phosphorylase(PYGL),glycogen synthase kinase(GSK-3),adipose triglyceride lipase(ATGL),which related to substance metabolism in liver and heart; as well as cytochrome C reductase(CCR),cytochrome C oxidase(COX),ATP synthase(ATPs),adenylate kinase(ADK),Na+-K+ ATPase and the levels of ATP、ADP、NAD+、NADH、NAD+/NADH,which related to energy metabolism.In this way,we can explore the influence on substance and energy metabolism of the cold and warm property of fraction of Mori Cortex.Results:Comparing with the normal group,the weight of Coptis chinensis Franch group and Scutellaria baicalensis Georgi group were reduced(P<0.05 or P<0.01)and there were no difference in other groups(P>0.05).The level of GCK,PFK-1,PGK,PK,ICD,CCR,COX,ATPs,ADK were significantly decreased(P<0.05 or P<0.01),acetyl-CoA,α-KGDHC,PYGL,ATGL were significantly increased(P<0.05 or P<0.01),the level of ATP was decreased without significantly and the levels of ADP、NAD+、NADH、NAD+/NADH had no significant difference(P>0.05)in the groups of Coptis chinensis Franch,Scutellaria baicalensis Georgi and Phellodendron chinense Schneid.The activity of GCK,PGK,CCR,COX,ATPs,ADK,ICD were significantly increased(P<0.05 or P<0.01)and acetyl-CoA,α-KGDHC,PYGL,ATGL were significantly decreased(P<0.05 or P<0.01),the level of ATP was increased without significantly and the levels of ADP、NAD+、NADH、NAD+/NADH had no significant difference(P>0.05)in the groups of Aconitum carmichaelii Debx,Zingiber offcinale Rosc and Zanthoxylum bungeanum.The activity of GCK、PFK-1、PGK、PK、PDH、ICD、GSK-3、CCR、COX、ATPs、ADK were significantly increased(P<0.05 or P<0.01)and α-KGDHC was significantly decreased(P<0.05)in the groups of 30% split fraction,fatty oil fraction of Mori Cortex,which showed the similar trend with cold medicine.Conclusion:The cold and warm property of Chinese traditional medicines can influence substance and energy metabolism of our body,which means the cold medicines can inhibit substance and energy metabolism and the warm medicines do the opposite,which is fit in with the new hypothesis that proposed by chief scientist Kuang Haixue professor.Overall,the groups of 30% split fraction,fatty oil fraction of Mori Cortex could inhibit substance and energy metabolism,which were similar with the cold medicines.We inferred that the property of these fractions maybe belong to the cold property.
