    目的:观察石斛合剂序贯治疗对db/db小鼠肝胰岛素信号通路蛋白基因表达的影响,探讨其调节血糖的分子机制。方法:选取12~14周龄雄性db/db小鼠,按空腹血糖及体质量随机分成模型组、二甲双胍组、石斛合剂序贯组(以下简称序贯组);选db/m为正常对照组。连续灌胃给药共6个循环(54 d)。观察各组治疗后各组空腹血糖(FBG)、口服葡萄糖耐量(OGTT)、糖化血清蛋白(GSP)、血清胰岛素(Ins)、三酰甘油(TG)及胆固醇(Tch),同时提取肝组织RNA,以逆转录PCR法检测胰岛素受体(InsR)、胰岛素受体底物1/2(IRS1/2)、PI3K mRNA表达水平变化。结果:与模型组比较,序贯组FBG、GSP和Ins显著下降(P<0.05),改善葡萄糖耐量(P<0.05~0.01);InsR、IRS1/2、PI3K等mRNA表达增加(P<0.05~0.01)。结论:复方石斛合剂序贯治疗能有效降低db/db小鼠的降低高胰岛素血症,降低空腹血糖及血脂,改善葡萄糖耐量,可能与增加InsR、IRS1/2、PI3K等基因表达有关。

    Abstract:

    To observe the effect of Shihu Compound (DC) sequential therapy on gene expression of protein of hepatic insulin signal pathways in db/db mice and to discuss the molecular mechanism of regulating blood sugar. Methods:Eighteen male db/db mice of 12~14 weeks old were randomly divided into model group, metformin group, and DC group according to FBG and body mass. Six db/m mice were taken as normal control group. They were given by continuous gavage for 6 circles (54 d). OGTT were measured at the end of the experiment. The concentrations of FBG, GSP, insulin, TG and Tch in sernm were measured, and the expression of insulin receptor, insulin receptor substrate 1/2 and PI3K in liver were assayed RT-PCR. Results:Compared with model group, FBG, Ins and GSP in DC group and metformin group decreased significantly (P<0.05), and glucose tolerance improved (P<0.05~0.01). The level of mRNA expression on InsR, IRS1/2 and PI3K were increased. Conclusion:Dendrobium Compound sequential treatment effectively reduced glucose and lipid, and improved glucose tolerance in db/db mice,which could be associated with increased the expressions of components in the insulin signal pathway, such as InsR, IRS1/2 and PI3K.
