栀子苷干预LPS诱导RAW264.7巨噬细胞炎性反应递质的研究

    目的:通过比较栀子苷水提液、栀子苷你标准品与栀子苷敲除液对LPS刺激巨噬细胞产生炎性反应的干预,探究栀子主要成分栀子苷的抗炎作用。方法:分别检测栀子苷水提液、栀子苷标准品、50%栀子苷敲除液对巨噬细胞的毒性;设立空白对照组、LPS模型组、栀子苷水提液组、栀子苷标准品组、50%栀子苷敲除液组、50%栀子苷回填组,检测各组对LPS刺激巨噬细胞后释放NO以及分泌IL-6和TNF-α的干预作用比较。结果:LPS刺激巨噬细胞后NO释放增多(P<10-4),而栀子苷水提液、栀子苷标准品、栀子苷敲除液均能降低LPS刺激巨噬细胞后NO的释放,其中栀子水提液、50%栀子苷敲除液以及50%栀子苷回填液的作用最强,且其作用差异无统计学意义(P<10-2)。而栀子苷标准品的作用相对较小(P<0.05)。LPS刺激巨噬细胞后,巨噬细胞分泌TNF-α与IL-6较空白对照组增加(P<10-4),栀子水提液干、栀子苷标准品、50%栀子苷敲除液以及含50%栀子苷的栀子水提液干预后,能降低上述细胞因子的分泌(P<0.05)。结论:栀子中栀子苷与其他成分均具有一定的抗炎作用,各成分之间的抗炎作用各有侧重。

    Abstract:

    To explore the anti-inflammatory effects of geniposide,a main component of Fructus Gardeniae,by comparing the intervention in aqueous extract of gardenoside,the standard product of geniposide and geniposide-knockout liquid on LPS-stimulated macrophage.Methods:Geniposide aqueous extract,geniposide standard product and 50%-geniposide-knockout liquid were used to detect macrophage cytotoxicity.The control group,LPS model group,and aqueous extract of geniposide liquid group,geniposide standard product group,50%-geniposide-knockout liquid group and 50%-geniposide-backfill liquid group were set up separately.The levels of NO,IL-6 and TNF-α in each group were detected in LPS-stimulated macrophages.Results:NO release was increased in LPS-stimulated macrophages (P<10-4),and aqueous extract geniposide,geniposide standard product,and geniposide-knockout liquid reduced the release of NO.Among them,the effects of geniposide aqueous extract,50%-geniposide-knockout liquid and 50%-geniposide-backfill liquid were the strongest (P<10-2),and there was no significant difference.The effects of geniposide standard product was relatively small (P<0.05).After stimulated by LPS,macrophages secreted more TNF-α and IL-6 than the control group (P<10-4),while geniposide aqueous extract,geniposide standard product,50%-geniposide-knockout liquid and 50%-geniposide aqueous extract could reduce the secretion of cytokines (P<0.05).Conclusion:Geniposide and other components of gardenia have a certain anti-inflammatory effect,and the anti-inflammatory effects of the various components have different emphases.
