    摘要:

    目的:观察肾脑复元汤对阿尔茨海默病大鼠海马区神经细胞的作用机制。方法:选取健康雄性SD大鼠120只,按完全随机法分为;假手术组(侧脑室注射3 μL蒸馏水),模型组(侧脑室注射3 μL β-淀粉样蛋白(Aβ)25-35+蒸馏水灌胃)、肾脑复元汤组(侧脑室注射3 μL β-淀粉样蛋白25-35+肾脑复元汤灌胃)及对照组(侧脑室注射3 μLβ-淀粉样蛋白25-35+盐酸多奈哌齐灌胃),各组均各灌胃1次,分别于7,14,21 d 3个时相处死。观察HE染色、tunnel染色及碱性成纤维生长因子(bFGF),脑源性神经营养因子(BDNF)的变化。结果:与模型组比较,中药组坏死程度较轻,且中药组(第14天)细胞凋亡程度较模型组减轻(P<0.05)。与模型组比较,中药组(第7天)bFGF,BDNF表达上升(P<0.05)。结论:肾脑复元汤对AD病大鼠海马区神经具有修复的作用,且发挥作用主要在后期。其机制与促进海马区BDNF及bFGF表达有关。

    Abstract:

    To observe the effects of Shennao Fuyuan Decoction on the influence and mechanism of nerve cell in rat hippocampus of Alzheimer's disease.Methods:A total of 120 healthy male SD rats were randomly divided into sham operation group(intracerebroventricular injection of 3 μL distilled water,),model group(intracerebroventricular injection of 3 μL Aβ25-35+distilled water administered intragastrically),Shennao Fuyuan Decoction group(intracerebroventricular injection of 3 μL Aβ25-35+Shennao Fuyuan Decoction administered intragastrically)and the control group(intracerebroventricular injection of 3 μL Aβ25-35+donepezil hydrochloride administered intragastrically).All the groups were administered intragastrically once and killed in 7,14,21 days.The changes of HE staining,tunnel staining and the levels of bFGF and BDNF protein were observed.Results:Compared with the model group,the degree of necrosis in the Chinese medicine group was mild,and the apoptosis in the traditional Chinese medicine group(14th day)was lower than that of the model group(P<0.01).Compared with the model group,the expression of bFGF and BDNF increased(P<0.05 or P<0.01)in the Chinese medicine group(7th day).Conclusion:Shennao Fuyuan Decoction is effective on the repairing nerve of rat hippocampus of AD disease,and plays a role in later period.Its mechanism is related to promoting the expression of BDNF and bFGF in hippocampus.
