    摘要:

    目的:评价利用超高压液相色谱/线性离子阱-静电场轨道阱高分辨质谱法(UPLC/LTQ-Orbitrapa MS)测定锁阳改善认知功能障碍有效部位的化学成分。方法:选取Acquity UPLC BEH C18(1.7 μm 2.1 mm×100 mm)色谱柱,柱温:30 ℃,流速:0.3 mL/min,进样量:5 μL,流动相为0.1%甲酸水(A)-乙腈(B)梯度洗脱。质谱采用电喷雾(ESI)离子源,在正负离子模式下采集数据。结果:共鉴别了21个化合物,包含6个潜在新化合物。鉴定的化合物包含核苷类、多酚类、黄酮苷类、氨基酸类、香豆素类化合物。结论:UPLC/LTQ-Orbitrapa MS方法能快捷、准确、较全面地鉴定锁阳有效部位的化学成分,为明确锁阳药效学物质基础及进一步提取分离提供科学依据。

    Abstract:

    To analyze and identify the chemical constituents from the active part of Herba Cynomorii using UPLC/LTQ-Orbitrapa MS (Ultra Performance Liquid Chromatography/Liner Ion Trap-Orbitrapa Mass Spectrometry).Methods:The analysis was performed on an Acquity UPLC BEH C18 reverse phase column (100 mm × 2.1 mm,1.7 μm).The injection volume was 5 μL.The flow rate was 0.3 mL/min gradient elution and column temperature was 30 ℃.The mobile phase consisted of acetonitrile (B) and 0.1% formic acid (A) was used as gradient elute.MS conditions were ESI (Electronic Spray Ion),positive and negative ion mode scanning.Results:After the data of mass spectra,pyrolysis rule and characteristic fragment ion information were analyzed,a total of 20 compounds were identified,including nucleosides,phenolic acids,flavonoid glycosides,Coumarins and amino acid.Conclusion:By using UPLC/LTQ-Orbitrapa MS method,the main chemical constituents from Herba Cynomorii can be accurately identified.
