Abstract:To observe the effects of Yougui Pills on gastrointestinal motility in rats with functional constipation,and to explore the concentration changes of intestinal neurotransmitters SP,NO,5-HT and VIP,as well as aquaporin 3(AQP3)in colon tissue.Methods:A total of 60 rats with functional constipation were randomly divided into blank group,model group and intervention group,with 20 rats in each group.The rats in the model group and the intervention group were given compound diphenoxylate suspension by gavage to prepare functional constipation models,and then the rats in the intervention group were given Yougui Pills suspension by gavage for 20 days.Gastric emptying rate and small intestinal propulsion rate in each group were calculated by gavage with activated carbon,levels of intestinal neurotransmitters SP,NO,5-HT and VIP in peripheral blood of rats were detected by enzyme-linked immunosorbent assay(ELISA),and expression of AQP3 in colon tissue of rats in each group was observed by Western blotting.Results:1)After the modeling,rats in the model group and the intervention group began to develop symptoms of listlessness,liking to lie down,loss of appetite,less fecal volume,dry stool,and even less urine in some rats.After the intervention,the above symptoms of rats were gradually improved,and symptoms of some rats disappeared. 2)The gastric emptying rate and small intestinal propulsion rate of the model group and the intervention group were significantly lower than those of the blank group,with statistically significant difference(P<0.05).The gastric emptying rate and small intestinal propulsion rate of the intervention group were significantly faster than those of the model group,with statistically significant difference(P<0.05). 3)In the model group and the intervention group,the levels of serum SP and VIP were decreased and the levels of NO and 5-HT were increased,compared with those in the blank group,with statistically significant difference(P<0.05),and those in the intervention group were superior to those in the model group,with statistically significant difference(P<0.05). 4)The expression of AQP3 in colon tissue of the model group and the intervention group was significantly higher than that of the blank group,with statistically significant difference(P<0.05),and the level of AQP3 in the intervention group was lower than that of the model group,with statistically significant difference(P<0.05).Conclusion:The occurrence of functional constipation may be related to abnormal intestinal neurotransmitter and aquaporin.Yougui Pills may be used to treat functional constipation by regulating the levels of SP,NO,5-HT,VIP and AQP3.
