Abstract:To explore the effects of Peiyuan Tongnao(PYTN)Capsules on working memory,content of Glu in hippocampus and NMDA Receptor 2B(NR2B)in rats with cerebral ischemia(CI).Methods:A total of 52 male SD rats were randomly divided into sham operation group(n=13),model group(n=13),nimodipine group(n=13)and PTNY group(n=13).The latter 3 groups were performed with bilateral carotid artery ligation.The nimodipine group and PTNY group were given corresponding drugs daily by gavage for 4 weeks according to body weight.The working memory of the rats was was tested by Morris water maze.The expression of Nissl body in hippocampus was observed by Nissl staining.Determination of Glu in hippocampus was conducted by High Performance Liquid Chromatography.The expression level of NR2B was determined by Western blot.Results:Morris water maze working memory escape latency:the sham operation group(DAY 1 versus DAY 2,DAY 1 versus DAY 3,P>0.05; DAY 1 versus DAY 4,P<0.05); the model group(DAY 1 versus DAY 2,DAY 1 versus DAY 3,P>0.05,DAY 1 versus DAY 4,P<0.05); the nimodipine group(DAY 1 versus DAY 2,DAY 1 versus DAY 3,P>0.05,DAY 1 versus DAY 4,P<0.05); the PYTN group(DAY 1 versus DAY 2,P>0.05,DAY 1 versus DAY 3,DAY 1 versus DAY 4,P<0.05).The content of Glu in hippocampus:the sham operation group was significantly different from the model group(P<0.05); the model group was significantly different from the PYTN group(P<0.05).The expression of NR2B:the sham group was significantly different from the model group,the nimodipine group and the PYTN group(P<0.01); the model group was significantly different from the nimodipine group(P<0.05); the model group was significantly different from the PYTN group(P<0.01).Conclusion:PYTN Capsules were helpful for improving working memory and protecting neurons in rats with cerebral ischemia,whose mechanism may associate with increasing Glu in hippocampus and the expression of NR2B in hippocampus.
