    摘要:

    目的:探究丹黄通脉方对颈动脉粥样硬化性狭窄模型大鼠炎性反应及脑血流量的影响。方法:将SPF级、雄性SD大鼠80只随机分为正常组、阳性对照组、模型组和观察组,每组20只。阳性对照组、模型组及观察组大鼠均采用高脂加高蛋氨酸复合饲料喂养联合颈总动脉球囊拉伤术法构建颈动脉粥样硬化性狭窄大鼠模型,正常组给予普通饲料喂养,且不进行颈总动脉球囊拉伤术。造模成功后,阳性对照组大鼠给予辛伐他汀2.5 mg/kg灌胃给药,观察组大鼠给予丹黄通脉方20 g/kg(浓度:10 mL/kg)灌胃给药,正常组与模型组则灌胃等量生理盐水,2次/d,各组均连续给药8周。干预结束后,采用全自动生化分析仪检测各组大鼠血清中血脂四项总胆固醇(TC),三酰甘油(TG),低密度脂蛋白胆固醇(LDL-C)及高密度脂蛋白胆固醇(HDL-C)水平;采用酶联免疫吸附试验法(ELISA)检测各组大鼠血清肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、C反应蛋白(CRP)及同型半胱氨酸(Hcy)水平;HE染色观察大鼠颈动脉病理学形态变化;采用激光多普勒血流仪测定各组大鼠脑血流量。结果:与正常组比较,模型组和观察组TC、TG、LDL-C水平,TNF-α、IL-6、CRP及Hcy水平显著升高,而HDL-C显著降低;但与模型组比较,阳性对照组和观察组TC、TG、LDL-C水平,炎性反应递质TNF-α、IL-6、CRP及Hcy水平显著降低,而HDL-C显著升高,差异有统计学意义(P<0.05);与正常组比较,模型组和观察组大鼠局部脑血流量显著降低;与模型组比较,阳性对照组和观察组大鼠脑血流量明显升高,差异有统计学意义(P<0.05)。结论:丹黄通脉方可有效降低颈动脉粥样硬化性狭窄大鼠的血脂,具有抗炎作用,可降低炎性递质水平,可增加脑血流量。

    Abstract:

    To investigate the effects of Danhuang Tongmai Recipe on inflammatory response and cerebral blood flow in rats with carotid atherosclerotic stenosis. Methods:A total of 80 SPF and male Sprague-Dawley rats were randomly divided into a normal group (n=20), a positive control group (n=20), a model group (n=20) and an experimental group (n=20). Rats in the positive control group, the model group and the experimental group were treated with high-fat plus methionine compound feed combined with carotid artery balloon injury to construct a rat model of carotid atherosclerotic stenosis. The normal group was fed with normal feed and no ballooning of the common carotid artery was performed. After successful modeling, rats in the positive control group were given simvastatin 2.5 mg/kg by intragastric administration, and rats in the experimental group were administered with Danhuang Tongmai Recipe 20 g/kg (concentration:10 mL/kg). The normal group and the model group were given the same amount of normal saline twice a day, and each group was continuously administered 8 weeks. After the intervention, the levels of TC, TG, LDL-C and HDL-C in the serum of the rats in each group were detected by automatic biochemical analyzer; The levels of TNF-α, IL-6, CRP and Hcy in the serum of each group were detected by ELISA; HE staining was used to observe the pathological changes of carotid artery in rats; The cerebral blood flow of each group was measured by laser Doppler flowmetry. Results:Compared with the normal group, the levels of TC, TG, LDL-C, TNF-α, IL-6, CRP and Hcy in the model group and experimental group significantly increased, while HDL-C significantly reduced; Compared with the model group, TC, TG, LDL-C levels, inflammatory factors TNF-α, IL-6, CRP and Hcy levels significantly reduced in the positive control group and the experimental group, while HDL-C significantly increased (P<0.05); At the same time, compared with the normal group, the cerebral blood flow significantly reduced in the model group and the experimental group; Compared with the model group, the cerebral blood flow of the positive control group and the experimental group significantly increased (P<0.05). Conclusion:Danhuang Tongmai Recipe can effectively reduce the blood lipids of rats with carotid atherosclerotic stenosis, with anti-inflammatory effects, which can reduce the level of inflammatory mediators. At the same time, it can also increase cerebral blood flow, which is worthy of clinical recommendation.
