To investigate the effects of Jinshuiqing Mixture on the expression of TGF-β1,CTGF,VEGF protein and mRNA in IgA nephropathy rats.Methods:A total of 40 4-week-old SPF SD rats were randomly divided into a normal groups,a model group,treatment group 1,treatment group 2 and treatment group 3.Rat IgA nephropathy model was copied.Based on the model group,the treatment group was given Jinshuiqing Mixture by gavage once per day in the 5th week,the 7th week and the 9th week respectively till the end of 12th week.Pathological changes in kidney were observed by HE staining and PAS staining.The protein expression of TGF-β1,CTGF and VEGF was detected by Western blot,and the expression of TGF-β1,VEGF and CTGF and mRNA was detected by RT-PCR.Results:In the normal group,the renal tissue structure was complete and clear.In the model group,mesangial area was widened,and mesangial cells and mesangial matrix were moderately to severely proliferated.The degree of lesions in the treatment group was lighter than that in the model group.And the improvement of treatment group 1 was the most obvious one.Compared with the normal group,the expression of TGF-β1,CTGF and VEGF protein in the model group increased significantly and the difference was statistically significant (P<0.05); compared with the model group,the expression in the treatment group decreased greatly and the difference was statistically significant (P<0.05); compared among the treatment groups,and the difference was statistically significant (P<0.05); compared with the normal group,the expression of TGF-β1,CTGF,VEGF mRNA in the model group was significantly higher,and there was significant difference (P<0.05); compared with the model group,the expression in the treatment group was lower,and the difference was statistically significant (P<0.05); compared among the treatment groups,and the difference was statistically significant (P<0.05).Conclusion:Jinshuiqing Mixture can significantly improve the renal damage of IgA nephropathy rats,and early treatment improvement is most obvious.Its mechanism may be down regulating the expression of TGF-β1,CTGF,VEGF protein and mRNA.
