Abstract:To investigate the effects of Sini Powder on goblet cells and the expression of mucin 2 (MUC2) in the duodenum of functional dyspepsia (FD) rats.Methods:A total of 30 SD rats were randomly divided into 3 groups with 10 rats in each group,including a control group,a model group and a Sini Powder treatment group.The FD animal model was induced by intragastric juvenile iodoacetamide gavage combined with tail clip stress stimulation.Normal group and model group were given intragastric saline.The Sini Powder treatment group rats was given Chinese medicine decoction of Sini Powder for intragastric administration.The local duodenal tissue of rats was taken for HE staining to observe the basic morphology of the duodenal tissue,for AB-PAS staining to observe the number of goblet cells,for immunohistochemistry (IHC) to observe the expression of MUC2 protein,for quantitative PCR (qPCR) detection of MUC2 mRNA expression.Results:HE staining showed that the basic morphology of the duodenum of rats was normal in the 3 groups.Compared with the normal group,the number of duodenal goblet cells in the model group was significantly reduced; MUC2 mRNA and immunopositive expression were significantly lower than the control group (P<0.01,P<0.01),the difference was statistically significant.Compared with the model group,the above indexes of the Sini Powder treatment group were significantly higher than the model group (P<0.01,P<0.05),the difference was statistically significant.Conclusion:Sini Powder can restore mucus barrier by increasing the number of goblet cells in duodenum,regulating the expression of MUC2 so as to treat FD.
