Abstract:To explore the therapeutic effect of Bushen Huoxue decoction on osteonecrosis of the femoral head and its effect on Wnt-β catenin signal pathway. Methods:A total of 45 SD rats were randomly divided into blank group (n=15), model group (n=15) and traditional Chinese medicine group (n=15). The animal model of steroid-induced osteonecrosis of the femoral head was made by methylprednisolone sodium succinate. the rats in the traditional Chinese medicine group were fed with Bushen Huoxue decoction once a day, and the rats in the blank group and model group were fed with normal saline for 6 weeks. The changes of femoral morphology, bone mineral density, pathology of femoral head, serum calcium, phosphorus, Wnt10b and β-Catenin in each group were compared. Results:1) the density of the femoral head of the model group was different, the femoral head was rough and the trabeculae were blurred. The number of trabeculae of the femoral head in the traditional Chinese medicine group was more than that in the model group, and it was relatively clear, and the femoral head in the traditional Chinese medicine group was smoother than that in the model group. 2) the bone mineral density (BMD), serum calcium and phosphorus levels, Wnt10b and β-Catenin protein in femoral head tissue of rats in model group and traditional Chinese medicine group were all decreased significantly (P<0. 05). It was higher in the traditional Chinese medicine group than that in the model group (P<0. 05). 3) there were obvious pathological changes in the cartilage of the femoral head in the model group, the structure of the femoral head in the traditional Chinese medicine group was still intact, the empty lacunae was less than that in the model group, and the arrangement of chondrocytes was irregular. Conclusion:Bushen Huoxue decoction has protective effect on osteonecrosis of femoral head, and its mechanism is related to Wnt-β catenin signal pathway.
