Abstract:To analyze the mechanism of Huoxue Jiangtang Drink on diabetic cardiomyopathy from the perspective of mitochondrial autophagy based on the animal models of diabetic cardiomyopathy intervened by Huoxue Jiangtang Drink.Methods:Male Wistar rats were randomly divided into a normal group,a model group,a rosiglitazone group and a Huoxue Jiangtang Drink group.The last 3 groups used STZ-induced diabetic rat models.Huoxue Jiangtang Drink group was treated with 2.96 g/(kg·d) intragastric administration,and the rosiglitazone group was treated with 4 mg/(kg·d) intragastric administration.After 8 weeks,total cholesterol,triglyceride,creatine kinase and lactate dehydrogenase were tested,echocardiography was conducted,myocardial tissue was collected,mitochondrial membrane potential was detected by flow cytometry.Myocardial tissue was collected,mitochondrial membrane potential was detected by flow cytometry,and the protein levels of Pink1,Parkin,Mfn1,Mfn2,and Beclin1 were detected by Western-blot.Results:Compared with the normal group,the model group had a significant increase in CK,LDH,TC,TG,a decrease in cardiac function,a remarkable increase in the ratio of membrane potential change,and a downtrend of the protein level of Pink1、Parkin、Mfn1、Mfn2、Beclin1.However,the Huoxue Jiangtang Drink group and the rosiglitazone group got a decrease in CK,LDH,TC,TG,an improvement of cardiac function,a decrease in the ratio of membrane potential changem,and a notable increase in the protein level of Pink1,Parkin,Mfn1,Mfn2,Beclin1.Conclusion:Huoxue Jiangtang Drink can significantly reduce the levels of CK,LDH,TC,TG and improve the cardiac function of diabetic cardiomyopathy rats.Futhermore,it can promote mitochondrial autophagy and protect myocardial cells by up-regulating the expression of PINK1,Parkin,Mfn1,Mfn2 and Beclin 1.
