    摘要:

    目的:研究白杨素对心肌缺血再灌注损伤(MI/RI)大鼠的作用,同时观察其对心肌凋亡相关蛋白Bax和Bcl-2表达的影响。方法:SPF级SD大鼠60只适应性喂养2周后,按体质量大小依次分为假手术组、模型组、阳性药物雷米普利组、白杨素低、中和高剂量组,每组10例,疗程2周,后采用可逆左冠状动脉前降支结扎法建立MI/RI模型,假手术组只穿线但不结扎。手术后收集血液检测血清肌酸激酶同功酶(CK-MB)、乳酸脱氢酶(LDH)、白细胞介素(Interleukin,IL)-6、超氧化物歧化酶(SOD)和丙二醛(MDA)含量,TTC法评估心肌梗死面积,HE染色观察心肌病理学,Western Blot检测心肌Bax和Bcl-2蛋白表达特点。结果:与假手术组比较,模型组心肌梗死面积明显增加(P<0.05),心肌纤维断裂,与模型组比较,白杨素3个剂量组和雷米普利组心肌梗死面积降低(P<0.05),心肌细胞形态改善;与假手术组比较,模型组血清CK-MB、LDH、IL-6和MDA增加(P<0.05),SOD降低(P<0.05),与模型组比较,白杨素3个剂量组和雷米普利组CK-MB、LDH、IL-6和MDA降低(P<0.05),SOD升高(P<0.05);与假手术组比较,模型组心肌Bax蛋白表达明显增加(P<0.05),Bcl-2表达降低(P<0.05),与模型组比较,白杨素3个剂量组和雷米普利组Bax蛋白表达降低(P<0.05),Bcl-2上调(P<0.05)。结论:白杨素对MI/RI大鼠血清心肌酶CK-MB和LDH,炎性因子IL-6,氧化和抗氧化指标MDA和SOD均有调节作用,同时可降低心肌梗死面积,其作用机制可能与白杨素能够调节Bcl-2和Bax蛋白表达有关。

    Abstract:

    To study the therapeutic effects of chrysin on MI/RI rats,and to observe the proteins expression of Bax and Bcl-2.Methods:According to weight,A total of 60 SD rats after adaptive feeding for 2 weeks were divided into a sham operation group,a model group,a positive drug ramipril group,chrysin low,medium and high dose groups,n=10.The course of treatment was 2 weeks.Later,the MI/RI model was established by reversible left anterior descending coronary artery ligation.The sham operation group only threaded but not ligated.Later,the MI/RI model was established by reversible left anterior descending coronary artery ligation.The sham operation group only threaded but not ligated.After the operation,blood was collected for serum creatine kinase isoenzyme(CK-MB),lactate dehydrogenase(LDH),interleukin(IL)-6,superoxide dismutase(SOD)and malondialdehyde(MDA)content.TTC method was used to evaluate myocardial infarction area,HE staining was used to observe myocardial pathology.Western Blot was used to detect myocardial Bax and Bcl-2 protein expression characteristics.Results:Compared with the sham group,the infarct size in the model group was increased(P<0.05),the myocardial fibers were broken,compared with the model group,the myocardial infarct size was improved in the 3 dose groups of Chrysin and Ramipril group(P<0.05).Cardiomyocyte morphology improved; compared with the sham group,serum CK-MB,LDH,IL-6 and MDA increased in the model group(P<0.05),SOD decreased(P<0.05).Compared with the model group,CK-MB,LDH,IL-6 and MDA in the 3 doses of chrysin and ramipril group decreased(P<0.05),and SOD decreased(P<0.05); Compared with the sham operation group,the expression of Bax protein in the myocardium of the model group was significantly increased(P<0.05),and the expression of Bcl-2 was decreased(P<0.05).Compared with the model group,the three-dose group of chrysin and expression Bax protein in Puri group was reduced(P<0.05),and Bcl-2 was up-regulated(P<0.05).Conclusion:Chrysin has a regulatory effect on serum myocardial enzymes CK-MB and LDH,inflammatory factor IL-6,oxidation and antioxidant indicators MDA and SOD in MI/RI rats,and can reduce the area of myocardial infarction.Its mechanism of action may be related to that of chrysin regulating Bcl-2 and Bax protein expression.
