To explore the effects of Yuzhi Zhixue Granules on the signal pathway of phosphatidylinositol 3-kinase/protein kinase B (PI3K/Akt) and the volume of ectopic endometrium in rats with endometriosis.Methods:The rat model of endometriosis was established and randomly divided into a model group,a low,middle,high dose Yuzhi Zhixue Granules group and a gestrinone group,with 12 rats in each group.Another 12 were set as sham operation group,the rats were killed after treatment,the volume of ectopic endometrium of rats was measured,the microvessel density (CD31 positive cell expression) was detected by immunohistochemistry,the levels of VEGF,MMP-9,IL-6 and TNF-α in serum were detected by enzyme linked immunosorbent assay (ELISA); and the expression of PI3K/Akt pathway related proteins was detected by Western blot.Results:Compared with the sham operation group,the proportion of CD31 positive cell,levels of VEGF,MMP-9,IL-6 and TNF-α in serum,the p-PI3K/PI3K and p-Akt/Akt in the ectopic endometrium tissues of the model group were all higher (P<0.05); Compared with the model group,the low,medium and high doses of Yuzhi Zhixue Granules and the ectopic endometrial volume of gestrinone rats,the proportion of CD31 positive cells,serum VEGF,MMP-9,IL-6 and TNF-α level,p-PI3K/PI3K and p-Akt/Akt of ectopic endometrial tissue were all decreased (P<0.05),and there was a dose-dependent relationship between Yuzhi Zhixue Granule groups (P<0.05); there was no significant difference between high dose Yuzhi Zhixue Granule group and gestrinone group (P>0.05).Conclusion:Yuzhi Zhixue Granule can inhibit the angiogenesis of endometrium,reduce inflammation and reduce the volume of endometrium,which may be achieved by blocking PI3K/Akt signal transduction.
