Abstract:To explore the mechanism of Edible Tulip-Bulbus Fritillariae Thunbergii on in the treatment of breast cancer based on network pharmacology.Methods:The active components and targets were collected from Traditional Chinese Medicine Systems Pharmacology Datebase Analysis Platform (TCMSP) database,and The active ingredients and targets of Edible Tulip-Bulbus Fritillariae Thunbergii were collected.We used Uniprot database to select the corresponding drug-action protein name corresponding to the gene name,constructed a “drug-active ingredient-target” network diagram through cytoscape 3.7.2,and searched the disease database (GeneCards) and the online human Mendelian Inheritance database (Online Mendelian Inheritance in Man,OMIM) to obtain breast cancer related targets.We used cytoscape 3.7.2 to construct a protein-protein interaction (PPI) network of “active ingredients and breast cancer intersection targets”,and screened PPI network nodes to obtain key targets; used Metascape database to perform gene ontology for key targets (GO) functional analysis and Kyoto Encyclopedia of Genes and Genome (KEGG) pathway enrichment analysis.We used cytoscape 3.7.2 to construct a protein-protein interaction (PPI) network of “active ingredients and breast cancer intersection targets”,and screened PPI network nodes to obtain key targets; used Metascape database to perform gene ontology for key targets (GO) functional analysis and Kyoto Encyclopedia of Genes and Genome (KEGG) pathway enrichment analysis.Results:The study obtained 10 important active ingredients from the Edible Tulip-Bulbus Fritillariae Thunbergii drug pair,including β-sitosterol,stigmasterol,2-methoxy-9,10-dihydrophenanthrene-4,5-diol,Gerberol and others acting on 71 key targets such as FUS and RPL24.GO enrichment analysis identified 31 items,including 3 cell components,8 molecular functions,and 20 biological processes.In addition,KEGG enrichment pathway analysis identified a total of 14 enrichment pathways.Conclusion:The effects of Edible Tulip-Bulbus Fritillariae Thunbergii drug pair on breast cancer reflects the characteristics of multi-component,multi-target,and multi-pathway of Chinese medicinal.Its mechanism of action may be through regulation of cancer pathways,regulation of signal pathways,regulation and mediation of cell maturation and Change the cell cycle and other aspects to play a role in the treatment of breast cancer.This research provides ideas and references for further research.
