Abstract:To explore the mechanism of Xinglou Chengqi Decoction in the treatment of acute cerebral infarction based on network pharmacology.Methods:Pharmacology database and analysis platform of traditional Chinese medicine system platform (TCMSP) combined with literature retrieval was used to obtain the effective drug components of Xinglou Chengqi Decoction; SwissTargetPrediction database was used to predict drug components related targets; OMIM and GeneCards database were used to obtain the disease-related targets of acute cerebral infarction; we drew a Venn diagram to get the potential target of Xinglou Chengqi Decoction in the treatment of acute cerebral infarction.STRING database and Cytoscape 3.8.0 software were used to construct potential target protein interaction network and drug-component-disease-target network; GO and KEGG enrichment analysis of potential targets were carried out using David 6.8 database.Results:Through TCMSP and related literature search,47 effective components of Xinglou Chengqi Decoction were obtained,including apigenin,vanillin,cholic acid,etc.A total of 85 potential targets of Xinglou Chengqi Decoction in treating acute cerebral infarction were obtained,including STAT3,ESR1,CCNB1,etc.A total of 225 functional items of GO enrichment analysis were obtained,including 132 biological process (BP),34 cell components (CC) and 59 molecular function (MF).There were 20 KEGG signaling pathways,including arachidonic acid metabolism,bile secretion,cAMP signaling pathway etc.Conclusion:Based on network pharmacology,the mechanism of treatment of acute cerebral infarction by Xinglou Chengqi Decoction is explained from the molecular level,which may be used to inhibit oxidative stress,anti-inflammatory,regulate energy metabolism,anti-apoptosis,etc,and provide clues and basis for subsequent experimental research and development of new drugs.
