To investigate the effect of Chaihu Longgu Muli Decoction(CLMD) on the expression of cyclooxygenase-2(COX-2)/prostaglandin E2(PGE2) in hippocampal astrocytes in mice induced by pentylenetetrazol(PTZ).Methods:Hippocampal astrocytes of mice were separated and cultured.The expression of glial fibrillary acidic protein(GFAP) was identified by immunofluorescence.The cells were randomized into control group,PTZ group,PTZ+low-dose CLMD(10 mg/L) group,and PTZ+high-dose CLMD(20 mg/L) group.The concentration of COX-2 and PGE2 in cell culture was detected by enzyme-linked immunosorbent assay(ELISA),and the expression of COX-2 and prostaglandin E receptor 2(PTGER2) was measured by Western blotting.Results:Immunofluorescence suggested the expression of GFAP in astrocytes.The levels of COX-2 and PTGER2 were significantly increased and the concentration of COX-2 and PGE2 was significantly raised in PTZ group compared with those in the control group(P<0.01).The levels of COX-2 and PTGER2 were significantly decreased and the concentration of COX-2 and PGE2 was also significantly reduced in PTZ+low-dose CLMD group and PTZ+high-dose CLMD group compared with those in the PTZ group(P<0.01).The effect of high-dose CLMD was more obvious(P<0.01).Conclusion:CLMD can inhibit COX-2/PGE2 inflammatory reaction pathway in PTZ-induced astrocytes.
