    摘要:

    目的:建立同时测定大鼠血浆样品中乌头碱、次乌头碱、苯甲酰乌头原碱含量的液相色谱-串联质谱(LC-MS/MS)方法,并用于制草乌给药后,3种成分在大鼠体内的药代动力学研究。方法:以小檗碱为内标,采用Shim-pack Velox SP-C18(100 mm×2.1 mm,2.7 μm)色谱柱,流动相为水-甲醇-乙腈(0.05%甲酸),流速0.3 mL/min,质谱采用电喷雾离子源(ESI),正离子模式,选择反应监测(MRM)方式。血浆样品采用有机试剂沉淀蛋白法。结果:乌头碱、次乌头碱、苯甲酰乌头原碱成分的线性关系良好(r≥0.999 0),日内和日间精密度符合生物样品分析要求,相对标准偏差(RSD)均小于15%。结论:此方法灵敏度高、专属性强,适用于测定乌头碱、次乌头碱、苯甲酰乌头原碱的浓度以及药代动力学研究。

    Abstract:

    To establish a liquid chromatography coupled with tandem mass spectrometry(LC-MS/MS) method for simultaneous determination of aconitine,hypaconitine and benzoylaconitine in rat plasma,and to study the pharmacokinetics of the three components in rats after administration of Radix Aconiti Kusnezoffii Praeparata.Methods:Berberine was used as internal standard,and chromatographic separation was performed on a Shim-pack Velox SP-C18(100×2.1 mm,2.7 μm) with water-methanol-acetonitrile(0.05% formic acid) as the mobile phase at the flow rate of 0.3 mL/min.The mass spectrometry was carried out by electrospray ionization(ESI),positive ion mode and multiple reaction monitoring(MRM).The plasma samples were precipitated by organic reagent.Results:The linear relationship of aconitine,hypaconitine and benzoylaconitine was good(r≥0.999 0).The intra-day and inter-day precision met the requirements of biological sample analysis,with RSD less than 15%.Conclusion:This method is highly sensitive and specific,and it is suitable for the determination of aconitine,hypaconitine and benzoylaconitine and the study of the pharmacokinetics.
